摘  要:以珙桐冬芽、腋芽、未成熟种子去胚乳及带胚乳的胚、成熟种子去胚乳及带胚乳的胚为外植体,对珙桐愈伤组织诱导及分化进行了研究。结果表明:珙桐未成熟种子去胚乳的幼胚是珙桐极佳的外植体,因其初代培养时不褐化;添加3种细胞分裂素6-BA、KT、ZT及较高浓度的生长素2,4-D的条件下幼胚分化形成愈伤组织;添加细胞分裂素6-BA和较低浓度的生长素NAA的条件下幼胚发育成珙桐完整小植株。继代培养阶段小植株茎段增殖和愈伤组织芽诱导分化的激素配比则有待进一步摸索。In this paper,callus induction and differentiation of Davidia involucrata were studied by using winter bud,axillary bud and embryo with endosperm and without endosperm of immature seeds and mature seeds as explants.The results showed that young embryo without endosperm of immature seeds was an excellent explant because it didn’t brown in the initial culture stage.It produced callus through differentiation in the medium supplemented with three kinds of cytokinin 6-BA,KT,ZT and a higher concentration of auxin 2,4-D,and it grew into an intact plants in the medium supplemented with cytokinin 6-BA and a lower concentration of auxin NAA.The hormone ratios of stem segment proliferation and callus bud induced differentiation need for further exploration in the subculture stage.
