摘  要:为探究食淀粉乳杆菌对轮状病毒感染中NSP1及NF-κB通路mRNA表达的影响,将食淀粉乳杆菌及其分泌上清分别作用于轮状病毒感染的乳鼠和IPEC-J2细胞,采用荧光定量PCR技术测定各组NSP1和IκB/NF-κB通路的mRNA水平变化。体内与体外试验结果显示,食淀粉乳杆菌及其分泌上清可显著下调NSP1 mRNA的表达(P<0.05)。体外试验中,预防性干预组(BI组)IκB、NF-κB整体趋势相似,与正常对照组相比NF-κB mRNA上调显著(P<0.05),与轮状病毒组(RV组)和治疗性干预组(AI组)相比表达相对平稳,IκB mRNA在第4到6小时上调显著(P<0.05);体内试验中,在乳鼠感染轮状病毒的3 d内BI组NF-κB mRNA显著上调且与RV组和AI组差异显著(P<0.05);第3天时BI组IκB mRNA水平上调高于RV组和AI组且差异显著(P<0.05),RV组和AI组无显著性差异(P>0.05)。结果表明,食淀粉乳杆菌及其分泌上清能够有效下调猪轮状病毒NSP1的mRNA表达,在激活IκB/NF-κB通路、提高机体先天性免疫方面具有积极作用,而且预防作用优于治疗作用。To investigate the effect of Lactobacillus amylovorus on expression of NSP1 and IκB/NF-κB pathway mRNA in rotavirus(RV)infection,L.amylovorus and its secreted supernatant were applied to RV-infected suckling mice and IPEC-J2 cells respectively.Fluorescence quantitative PCR technique was used to determine the m RNA levels of NSP1 and IκB/NF-κB pathway in each group.Results showed that L.amylovorus and its secreted supernatant could reduce the expression of NSP1 mRNA(P<0.05).In the cell test,the trend of IκB and NF-κB in the intervention before RV infection group(BI group)was similar.NF-κB mRNA was significantly higher than the control group(P<0.05).Compared with RV group and AI group,its expression was relatively stable.IκB mRNA was significantly up-regulated from 4 h to 6 h(P<0.05).In the mice test,the NF-κB mRNA level of the BI group was markedly up-regulated and significantly different from the RV group and the AI group within 3 days of RV infection in the suckling mice(P<0.05).On the third day,the level of IκB mRNA in the BI group was higher than RV group and AI group and had significant difference(P<0.05)and RV group and AI group had no significant difference(P>0.05).The results indicated that L.amylovorus and its secreted supernatant can effectively reduce the expression of the RV NSP1 at the mRNA level and it plays a positive role in activating the IκB/NF-κB pathway and improving the body’s innate immunity.Preventive effect is better than therapeutic effect.
