摘 要:目的探讨白毛藤多糖对骨肉瘤细胞增殖、迁移、侵袭的影响及其机制。方法选取海南省文昌市人民医院骨科2017年1月至2019年1月接收的骨肉瘤患者30例,经病理检测确诊为骨肉瘤,将骨肉瘤细胞分为对照组、白毛藤多糖低、中、高浓度组、pcDNA3.1组、pcDNA3.1-Ebp11组、白毛藤多糖+si-NC组、白毛藤多糖+si-Ebp1组。细胞计数试剂盒(CCK-8)检测细胞增殖抑制率;蛋白质印迹(Western blot)法检测蛋白表达;Transwell检测细胞迁移和侵袭;实时定量反转录聚合酶链反应(RT-qPCR)检测ErbB3结合蛋白1(Ebp1) mRNA表达水平。两组比较行t检验,多组间比较采用单因素方差分析,组间两两比较采用LSD-t检验。结果白毛藤多糖低、中、高浓度处理的骨肉瘤细胞中细胞增殖抑制率[(16.39±1.68)%、(37.25±3.44)%、(58.14±5.71)%比(0.00±0.02)%,F=485.673,P<0.05]显著升高,细胞迁移数[(115.69±10.25)、(81.29±8.34)、(62.33±6.24)个比(136.25±11.21)个,F=117.477,P<0.05]、侵袭数[(87.32±8.33)、(69.65±6.58)、(51.26±5.22)个比(113.54±9.87)个,F=106.720,P<0.05]显著降低,细胞周期蛋白D1(Cyclin D1)(0.49±0.05、0.37±0.04、0.24±0.03比0.62±0.06,F=110.791,P<0.05)、基质金属蛋白酶(MMP)-2(0.58±0.05、0.44±0.04、0.32±0.03比0.71±0.07,F=104.091,P<0.05)、MMP-9蛋白相对表达水平显著降低(0.46±0.04、0.34±0.03、0.21±0.02比0.59±0.05,F=176.444,P<0.05),p21蛋白相对表达水平显著升高(0.49±0.04、0.63±0.05、0.76±0.07比0.31±0.03,F=135.364,P<0.05),Ebp1 mRNA(3.71±0.33、3.05±0.31、2.61±0.26比1.01±0.09,F=169.406,P<0.05)和蛋白相对表达水平显著升高(0.52±0.05、0.67±0.06、0.81±0.08比0.39±0.04,F=84.660,P<0.05)。骨肉瘤组织中Ebp1 mRNA(0.42±0.04比1.00±0.09,t=32.255,P<0.05)和蛋白相对表达水平显著降低(0.39±0.04比0.82±0.07,t=29.213,P<0.05)。过表达Ebp1抑制骨肉瘤细胞增殖、迁移和侵袭。抑制Ebp1表达逆转了白毛藤多糖对骨肉瘤U2OS细胞增殖、迁移和侵�Objective:To investigate the effect and mechanism of Solanum lyratum polysaccharide on the proliferation,migration and invasion of osteosarcoma cells.Methods:Thirty patients with osteosarcoma received from January 2017 to January 2019 in the Department of Orthopedics,Wenchang City People’s Hospital,Hainan Province were selected and diagnosed as osteosarcoma by pathological examination.The osteosarcoma cells were divided into control group,low,medium,and high concentration of Solanum lyratum polysaccharide,pcDNA3.1 group,pcDNA3.1-Ebp11 group,Solanum lyratum polysaccharide+si-NC group,Solanum lyratum polysaccharide+si-Ebp1 group.Cell counting kit 8(CCK-8)to detect cell proliferation inhibition rate;Western blotting method to detect protein expression;Transwell to detect cell migration and invasion;real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR)was used to detect the expression of ErbB3 binding protein 1(Ebp1)mRNA.The two groups were compared by t test,the comparison between multiple groups was by single factor analysis of variance,and the comparison between groups was by LSD-t test.Results:The inhibitory rate of cell proliferation[(16.39±1.68)%,(37.25±3.44)%,(58.14±5.71)%vs.(0.00±0.02)%,F=485.673,P<0.05]in osteosarcoma cells treated with low,medium and high concentration of Solanum lyratum polysaccharide were significantly increased,and the number of cell migration[(115.69±10.25),(81.29±8.34),(62.33±6.24)vs.(136.25±11.21),F=117.477,P<0.05],number of invasions[(87.32±8.33),(69.65±6.58),(51.26±5.22)vs.(113.54±9.87),F=106.720,P<0.05)]were significantly reduced,Cyclin D1(0.49±0.05,0.37±0.04,0.24±0.03 vs.0.62±0.06,F=110.791,P<0.05),matrix metalloproteinase(MMP)-2(0.58±0.05,0.44±0.04,0.32±0.03 vs.0.71±0.07,F=104.091,P<0.05),MMP-9(0.46±0.04,0.34±0.03,0.21±0.02 vs.0.59±0.05,F=176.444,P<0.05)protein relative expressions were significantly reduced,p21(0.49±0.04,0.63±0.05,0.76±0.07 vs.0.31±0.03,F=135.364,P<0.05)protein relative expression was significantly increase
