参附强心颗粒中红参提取工艺研究

摘  要:目的优选参附强心颗粒中红参提取工艺。方法通过水提取红参、水提醇沉以及不同醇度乙醇提取等7个不同工艺提取红参,所得到的提取物根据化学指标人参皂苷的含量以及离体心脏药理模型作用强弱两个方面综合评价,优选红参最佳提取工艺。结果在提取工艺方面,通过测定9种人参皂苷的含量,发现乙醇加热提取较水煎煮更有利于人参皂苷类成分的溶出。在对豚鼠离体心脏心功能影响方面,红参提取物有扩张冠脉的作用,并可显著提高冠脉流量。红参提取物在0.025 g生药/mL质量浓度下,与给药前相比冠脉流量升高,但心脏的收缩功能没有提高,甚至有部分提取物组的个别指标(如+dp/dtmax等)在给药后反而有所降低。红参70%和80%乙醇提取物对冠脉流量的提高最为明显,并显著优于水提物和90%乙醇提取物,收缩功能相关指标各组之间并无显著差别。结论化学指标和药理实验综合评价,70%和80%乙醇提取工艺效果最好,综合考虑工业化生产的易于实现性,70%乙醇提取工艺为参附强心颗粒中红参最佳提取工艺。Objective:To select the extraction process of Red ginseng in Shenfu Qiangxin Granules.Methods:Red ginseng was extracted by seven different processes,including water extraction,alcohol precipitation and alco⁃hol extraction with different alcohol degrees.According to the chemical index ginsenoside content and the effect of isolated heart pharmacological model,the best extraction process of red ginseng was optimized.Results:In terms of extraction process,by measuring the contents of 9 kinds of ginsenosides,it was found that ethanol extraction was more beneficial to the dissolution of ginsenosides than boiling water.In terms of its effect on the heart function of isolated guinea pig heart,red ginseng extract has the effect of expanding the coronary arteries and can significantly increase coronary flow.At a concentration of 0.025 g crude drug/mL,the red ginseng extract increased coronary flow compared with that before administration,but the heart′s systolic function was not improved.There were even individual indicators of some extract groups(such as+dp/dtmax,etc.).It decreased after administration.Red gin⁃seng 70%and 80%ethanol extracts had the most significant improvement in coronary flow,significantly better than water extracts and 90%ethanol extracts.There was no significant difference between groups of indexes relat⁃ed to contractile function.Conclusion:Comprehensive evaluation of chemical indexes and pharmacological experi⁃ments showed that 70%and 80%ethanol extraction process was the best.Considering the easy realization of indus⁃trial production,70%ethanol extraction process was the best extraction process of red ginseng in Shenfu Qiangxin Granules.
