摘 要:目的观察葛根素对百草枯中毒大鼠急性肾损伤(AKI)的保护作用及肾组织丝裂原活化蛋白激酶(MAPK)蛋白的影响。方法选30只雄性SD大鼠随机分为对照组、模型组和葛根素组。模型组和葛根素组腹腔注射百草枯(30 mg/kg),6 h后葛根素组腹腔注射葛根素(50 mg/kg),对照组和模型组腹腔注射等量生理盐水,持续3 d。在末次给药1 h后麻醉大鼠,进行心脏取血和分离肾脏,对肾组织进行病理学和肾功能检查,检测血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)水平,检测肾组织中活性氧自由基(ROS)水平及抗磷酸化P38抗体(p-P38)、磷酸化JNK抗体(p-JNK)和磷酸化ERK1/2(p-ERK1/2)水平。结果对照组未观察到损伤迹象;模型组出现肾小球出血,有炎性细胞浸润,肾小管可见蛋白管型和玻璃样变性;葛根素组肾小球基本正常,肾小管见少量蛋白管型和玻璃样变性,肾间质见炎细胞浸润和少量出血。与对照组比较,模型组和葛根素组血清中BUN、Scr、MDA、TNF-α和IL-1β及肾组织中ROS、pP38、p-JNK和p-ERK1/2水平均显著增加,SOD显著降低(P<0.05);与模型组比较,葛根素组血清中BUN、Scr、MDA、TNF-α和IL-1β及肾组织中ROS、p-P38、p-JNK和p-ERK1/2水平均显著降低,SOD显著增加(P<0.05)。结论葛根素对百草枯中毒大鼠AKI具有保护作用,其机制可能与抑制肾脏MAPK活性有关。Objective:To investigate the protective effect of Puerarin on acute renal injury induced by paraquat poisoning in rats and the effect on MAPK protein in renal tissue.Methods:30 male SD rats were divided into the control group,the model group and Puerarin group.The model group and Puerarin group were intraperitoneally in⁃jected with Paraquat(30 mg/kg),and Puerarin group was intraperitoneally injected with Puerarin(50 mg/kg)6 hours later.The control group and the model group were intraperitoneally injected with the same amount of saline for 3 days.One hour after the last administration,anesthetized rats were subjected to cardiac blood collection and kidney isolation.Pathological and renal function tests were performed on the kidney tissues.Serum levels of MDA,SOD,TNF-αand IL-1βwere measured.The levels of ROS and p-P38,p-JNK and p-ERK1/2 in the kidney tis⁃sues were also measured.Results:No damage was observed in the control group.The glomerular hemorrhage,in⁃flammatory cell infiltration,tubular protein tubular and vitreous degeneration were observed in the model group.The glomerulus was basically normal in Puerarin group;tubular protein tubular and vitreous degeneration were ob⁃served in a small amount;interstitial inflammatory cell infiltration and a small amount of hemorrhage were ob⁃served in Puerarin group.Compared with the control group,the levels of BUN,Scr,MDA,TNF-αand IL-1βin se⁃rum and ROS,p-P38,p-JNK and p-ERK1/2 in kidney tissue of the model group and Puerarin group were signifi⁃cantly increased,while the level of SOD was significantly decreased(P<0.05).Compared with the model group,the levels of BUN,Scr,MDA,TNF-αand IL-1βin serum of Puerarin group and ROS,p-P38,p-JNK and p-ERK1/2 in kidney tissue were significantly decreased,while the levels of SOD was significantly increased(P<0.05).Conclusion:Puerarin has protective effect on AKI in rats with Paraquat poisoning,and its mechanism may be related to the inhibition of renal MAPK activity.
