摘 要:为进一步挖掘小鼠高脂响应基因并阐明其分子机制,采用生物信息学方法分析正常和高脂诱导小鼠肝脏的基因表达谱芯片,鉴定出82个与小鼠高脂响应相关的差异表达基因(differentially expressed genes,DEGs)。对DEGs进行基因本体(gene ontology,GO)分析,结果显示这些DEGs主要参与了胆固醇运输、类固醇生物合成和脂质稳态等生物学过程。京都百科全书基因和基因组(Kyoto encyclopedia of genes and genomes,KEGG)分析结果表明,DEGs主要和系统性红斑狼疮、类固醇激素生物合成、胆固醇代谢等通路有关。对DEGs构建蛋白蛋白相互作用(protein-protein interaction,PPI)网络,通过cytoHubba计算得到Pcsk9、Lpl、Cd68、Saa3、Lgals3、Cxcl9、Orm2、Abcg5、H2-Ab1、Lyz2、Anxa5、Anxa2等12个关键基因(hub genes),其中,Pcsk9、Lpl、Abcg5为已报道的血脂代谢相关基因,其余9个为新发现的小鼠高脂响应基因。研究结果将有助于阐明小鼠高脂响应基因的机制,为血脂代谢相关分子机制研究提供理论基础。To further explore the high-fat response genes of mouse and clarify their molecular mechanisms,the gene expression profiling of normal and high-fat induced mouse liver were analyzed by bioinformatics methods.A total of 82 differentially expressed genes(DEGs)related to high-fat response of mouse were obtained.Gene ontology(GO)analysis of DEGs was carried out and the results showed that these DEGs mainly participated in cholesterol transport,steroid biosynthesis and lipid homeostasis.In addition,the analysis results of KEGG(Kyoto encyclopedia of genes and genomes)showed that these differentially expressed genes were mainly related to systemic lupus erythematosus,steroid hormone biosynthesis,cholesterol metabolism and other pathways.Furthermore,a PPI(protein-protein interaction)network was constructed for DEGs.Finally,twelve key genes(hub genes)including Pcsk9,Lpl,Cd68,Saa3,Lgals3,Cxcl9,Orm2,Abcg5,H2-Ab1,Lyz2,Anxa5,Anxa2 were calculated by cytoHubba.Among them,Pcsk9,Lpl,Abcg5 were reported lipid metabolism related genes,and the remaining nine genes were newly discovered mouse high-fat response genes.These results will be helpful to elucidate the mechanism of high-fat response gene in mice,and provide a theoretical basis for the study of molecular mechanisms related to lipid metabolism.
