祛浊化瘀中药对尿酸盐诱导中性粒细胞胞外诱捕网的影响

摘　　要：目的:探讨祛浊化瘀药对尿酸盐诱导中性粒细胞胞外诱捕网(NETs)治疗痛风性关节炎症的分子机制。方法:采用祛浊化瘀中药干预人中性粒细胞,采用免疫荧光染色观察NETs表达,Western blot检测细胞核NF-κB/P65表达,ELISA检测IL-1β含量。结果:祛浊化瘀中药干预后,NETs形成下降,高剂量组(5.36±0.91)ng/mL与对照组比较具有显著差异(P=0.005);NF-κBP65/GAPDH在高剂量为0.42±0.13,较对照组明显下降(P=0.000),在低剂量组为1.56±0.49,较对照组明显下降(P=0.015);高剂量组IL-1β为166.8±17.9pg/mL,较对照组明显下降(P=0.000)。结论:祛浊化瘀中药通过抑制NETs生成,下调NF-κB的表达,从而降低IL-1β的分泌,为中药治疗痛风提供了理论依据。Objective:Gout is a common inflammatory arthritis. Neutrophil Extracellular Traps(NETs) play an important role in gout inflammation. Removing turbidity and Promoting Blood Circulation have a good clinical effect in the treatment of gout inflammation, but the mechanism is unknown. This study will explain the molecular mechanism of Chinese medicine for promoting blood circulation and removing blood stasis in treating gout inflammation. Methods:Human neutrophils were treated with Chinese herbs for promoting blood circulation and removing blood stasis. The expression of NETs was observed by immunofluorescence staining. The expression of NF-κB/P65 was detected by Western blot. The content of IL-1β was detected by ELISA. Results:After intervention of Chinese herbs, the formation of NETs decreased, and the high dose group(5.36±0.91) ng/mL was significantly different from the control group(P=0.005). The high dose of NF-κB/P65/GAPDH was 0.42±0.13, significantly lower than the control group(P=0.000), 1.56±0.49 in the low-dose group, significantly lower than the control group(P=0.015);high-dose group IL-1β was 166.8±17.9 pg/mL, compared with the control group Significantly decreased(P = 0.000).Conclusion: Chinese medicine for Removing turbidity and Promoting Blood Circulation can inhibit the expression of NF-κB by inhibiting the production of NETs, thereby reducing the expression of IL-1β, and providing a theoretical basis for the treatment of gout with Traditional Chinese Medicine.
