摘 要:目的转化生长因子β(transforming growth factorβ,TGFβ)和结缔组织生长因子(CTGF)是肾小球硬化过程中的重要细胞因子,探究新一代血管紧张素转化酶抑制剂(angiotensin converting enzyme inhibitor,ACEI)-福辛普利(fosinopril,FOS)对大鼠肾小球系膜细胞(glomerular mesangial cells,GMC)中TGFβ 和CTGF 表达的影响。方法 建立体外培养大鼠肾小球系膜细胞,实验分为3 组:对照组(Ctrl组)、脂多糖组(LPS 组,内毒素)和福辛普利组(FOS 组),分别于6 h、12 h 和24 h 时间点观察酶联免疫吸附方法(ELISA)测定细胞上清液中TGFβ 和CTGF 的含量,荧光定量(RT-PCR)法检测TGFβ和CTGF 的mRNA 表达变化。结果 正常培养的大鼠肾小球系膜细胞均有微量的TGFβ 和CTGF 的分泌和mRNA 表达;脂多糖组在6 h、12 h 和24 h 时间点TGFβ 和CTGF 的分泌和mRNA 表达均高于对照组(均P < 0.01),福辛普利组TGFβ 和CTGF 的分泌和mRNA 表达均低于脂多糖组(均P < 0.01)。结论 内毒素可诱导大鼠肾小球系膜细胞分泌TGFβ 和CTGF 等细胞因子,福辛普利可抑制TGFβ 和CTGF 的分泌和mRNA 表达,为ACEI 类药物对肾脏保护作用提供实验依据。Objective Transforming growth factorβ(TGFβ)and connective tissue growth factor(CTGF)are important cytokines in the process of glomerulosclerosis.To explore the effect of a new generation of angiotensin converting enzyme inhibitor(ACEI),fosinopril(FOS)on the expressions of TGFβand CTGF in rat glomerular mesangial cells(GMC).Methods Rat mesangial cells were cultured in vitro.The cells were divided into three groups:control group,lipopolysaccharide group(LPS group,endotoxin group),and fosinopril group(FOS group).The contents of TGFβand CTGF in the supernatant of cells were measured by enzyme-linked immunosorbent assay(ELISA)at 6 h,12 h,and 24 h,respectively,the changes of mRNA expressions of TGFβand CTGF were measured by RT-PCR.Results In the normally cultured rat mesangial cells,the secretions and mRNA expressions of TGFβand CTGF were all traced;the secretions and mRNA expressions of TGFβand CTGF in the LPS group were higher than those in the control group at 6 h,12 h,and 24 h(all P<0.01),the secretions and mRNA expressions of TGFβand CTGF in the FOS group were lower than those in the LPS group(all P<0.01).Conclusion Endotoxin can induce glomerular mesangial cells to secrete TGFβand CTGF,fosinopril can inhibit the secretion and mRNA expressions of TGFβand CTGF,and provide experimental basis for ACEI drugs to protect kidney.
