摘  要:目的:探讨caspase-3抑制剂对顺铂诱导正常大鼠肾小管上皮细胞(normal rat kidney tubular epithelial cell,NRK-52E)损伤的影响以及可能的调控机制。方法:不同浓度的顺铂注射液和caspase-3抑制剂作用于NRK-52E细胞,用流式细胞术检测上述药物作用后的NRK-52E细胞的早凋亡率。结果:顺铂诱导后的NRK-52E细胞早凋亡率较对照组明显升高,NRK-52E细胞早凋亡率与顺铂浓度呈正相关,caspase-3抑制剂预处理组NRK-52E凋亡程度较顺铂组轻(P<0.01);且20μmol/L浓度的caspase-3抑制剂比10μmol/L浓度的caspase-3抑制剂对减轻顺铂诱导的NRK-52E细胞凋亡作用更佳。但随着顺铂浓度及诱导凋亡时间的增加,caspase-3抑制剂对减轻顺铂诱导的NRK-52E细胞凋亡作用有限。结论:顺铂会明显增加NRK-52E细胞早凋亡率,caspase-3抑制剂预处理能减轻顺铂诱导的NRK-52E细胞早凋亡率,caspase-3抑制剂对顺铂诱导NRK-52E损伤具有保护作用。Objective:To investigate the effect of caspase-3 inhibitor on cisplatin-induced normal rat kidney tubular epithelial cells(NRK-52E)injury and possible regulatory mechanisms.Methods:Different concentrations of cisplatin injection and caspase-3 inhibitor were applied to NRK-52E cells,and the rate of early apoptosis NRK-52E cells after the above drugs was detected by flow cytometry.Results:The rate of early apoptosis NRK-52E cells induced by cisplatin was significantly higher than that of the control group.The rate of early apoptosis NRK-52E cells was positively correlated with the concentration of cisplatin.The apoptosis of NRK-52E cells in the caspase-3 inhibitor pretreatment group was lighter than that of cisplatin group(P<0.01).And the concentration of caspase-3 at a concentration of 20μmol/L is better than that of a caspase-3 inhibitor at a concentration of 10μmol/L to alleviate cisplatin-induced apoptosis of NRK-52E cells.However,as cisplatin concentration and time to induce apoptosis increased,caspase-3 inhibitors had a limited effect on alleviating cisplatin-induced apoptosis of NRK-52E cells.Conclusion:Cisplatin can significantly increase the early apoptosis rate of NRK-52E cells.Pretreatment with caspase-3 inhibitor can attenuate the early apoptosis rate of NRK-52E cells induced by cisplatin.Caspase-3 inhibitor has protective effect on cisplatin-induced NRK-52E cells injury.
