摘 要:目的:研究长期大强度运动后心肌核因子κB(NFκB)和细胞间黏附分子-1(ICAM-1)的变化及其在运动性心肌纤维化中的作用。方法:72只雄性SD大鼠,随机分为安静对照组(Sed组,n=24)、中强度运动组(ME组,n=24)和大强度运动组(IE组,n=24),每组按照训练周期又分为8周、12周和16周组,各8只。安静对照组自由活动,中强度运动组和大强度运动组分别以速度15.2 m/min、坡度5°和速度28 m/min、坡度10°的条件每天运动1 h,每周运动5天。各组最后一次运动结束24小时后处死,迅速分离心脏取左、右心室。采用Western blot和RT-PCR分别检测NFκB、核因子κB抑制蛋白(IκB)和ICAM-1蛋白及m RNA表达的变化。结果:16周大强度运动后右心室NFκB/IκB m RNA水平显著高于安静对照组(P<0.05);8周(P<0.05)、12周(P<0.05)和16周(P<0.05)大强度运动后右心室NFκB/IκB蛋白表达水平显著高于安静对照组,而左心室NFκB/IκB蛋白表达水平在各个时间点的各组间没有显著性差异。16周大强度运动后右心室ICAM-1蛋白(P<0.01)和m RNA(P<0.05)表达水平显著高于安静对照组,而在8周和12周时各组间没有显著性差异;左心室ICAM-1蛋白表达水平在各个时间点的各组间没有显著性差异。结论:长期大强度运动后右心室心肌细胞中NFκB被激活且ICAM-1表达水平上调,表明长期大强度运动后右心室心肌细胞可能长期处于炎症状态,而炎症则可激活促纤维化信号通路导致右心室心肌纤维化。Objective To explore the effect of long-term intensive exercise on the myocardial nuclear factor κB(NFκB) and intercellular cell adhesion molecule-1(ICAM-1) and its role in exercise-induced myocardial fibrosis.Methods Seventy-two male Sprague-Dawley rats were randomly divided into a sedentary(Sed) group,a moderate exercise(ME) group and an intensive exercise(IE) group,each of 24. Rats in the Sed group were free of exercises,while those in ME and IE groups ran an hour on treadmill at 5° and 10° slopes at the speed of15.2 m/min and 28 m/min respectively every day,5 days per week. Twenty-four hours after the 8-,12-and16-week training,all rats were sacrificed and their both ventricles were isolated. Western blotting and RT-PCR were used to detect the changes of protein and gene expression of NFκB,IκB and ICAM-1,respectively.Results After 16-week intensive exercise,the average mRNA expression level of NFκB/IκB in the right ventricle was significantly higher than the control group(P<0.05),while 8-,12-and 16-intensive exercise,the protein expression level of NFκB/IκB in the right ventricle was significantly higher than the control group(P<0.05 for all). The average protein and mRNA expression of ICAM-1 in the right ventricle after 16 weeks of intensive exercise was significantly higher than the control group(P<0.01,P<0.05).Conclusion After long-term intensive exercise,the expression of NFκB is activated and ICAM-1 is up-regulated in the right ventricle,which suggests that prolonged inflammation might occur in the right ventricle,and contribute to the myocardial fibrosis.
