摘  要:目的观察A型肉毒毒素(BTX-A)对带状疱疹后神经痛(PHN)小鼠行为学反应的影响。方法选择造模成功的PHN小鼠36只,随机分为对照组、BTX-A组和背根神经节(DRG)/BTX-A组,每组12只,分别在疱疹局部注射0.9%氯化钠注射液1 ml、疱疹局部或第5腰椎DRG处注射BTX-A(2 U/ml)1 ml,1次/d,连续7 d。于注射前(Z0)和注射后第1、3、5、7天(Z1、Z3、Z5、Z7)检测各组小鼠的机械痛阈值和热痛阈值。结果Z0、Z1、Z3时,3组间机械痛阈值比较,差异均无统计学意义(均P>0.05)。Z5和Z7时,BTX-A组和DRG/BTX-A组机械痛阈值均明显高于对照组(均P<0.05),但BTX-A组、DRG/BTX-A组组间比较,差异无统计学意义(P>0.05)。Z0、Z1时,3组间热痛阈值比较,差异无统计学意义(P>0.05)。Z3、Z5和Z7时,BTX-A组和DRG/BTX-A组小鼠热痛阈值均明显低于对照组(均P<0.05),但BTX-A组、DRG/BTX-A组组间比较差异无统计学意义(P>0.05)。结论BTX-A可有效提高PHN小鼠机械痛阈值和降低PHN小鼠热痛阈值,为BTX-A临床治疗PHN提供了实验基础。Objective To investigate the effect of botulinum toxin A(BTX-A)on behavioral response of mice with postherpetic neuralgia(PHN).Methods PHN model was induced by subcutaneous injection of Herpes simplex virus-1(HSV-1)10μl(1×10^6 PFU/ml)in C57 mice.Thirty six PHN mice were randomly divided into three groups with 12 mice in each group.Mice in control group were injected with 1 ml normal saline into the local herpes,mice in BTX-A group and DRG/BTX-A were injected with 1 ml of BTX-A(2 U/ml)into the local herpes or dorsal root ganglion(DRG)at 5th lumbar vertebra once a day for 7 days,respectively.The mechanical pain threshold and thermal pain threshold were measured before injection(Z0)and on the 1st,3rd,5th,7th day(Z1,Z3,Z5 and Z7)after injection.Results There was no significant difference in mechanical pain threshold among the three groups at time points Z0,Z1 and Z3(all P>0.05).The mechanical pain threshold of BTX-A group and DRG/BTX-A group was significantly higher than that of control group at Z5 and Z7(all P<0.05),but there was no significant difference between BTX-A group and DRG/BTX-A group(P>0.05).There was no significant difference in thermal pain threshold among the three groups at time points Z0 and Z1(P>0.05).The thermal pain threshold of BTX-A group and DRG/BTX-A group was significantly lower than that of control group at Z3,Z5 and Z7(all P<0.05),but there was no significant difference between BTX-A group and DRG/BTX-A group(P>0.05).Conclusion BTX-A can effectively improve the mechanical pain threshold and reduce the thermal pain threshold of PHN mice.
