摘  要:目的:建立HPLC梯度洗脱法同时测定小儿葫芦散中天麻素、巴利森苷E、巴利森苷A和橙皮苷的含量。方法:采用Agilent Extend C18(200 mm×4.6 mm,5μm)色谱柱,柱温25℃;流动相为甲醇-0.1%甲酸溶液,梯度洗脱,流速0.8 mL/min;检测波长分别为220 nm(检测天麻素、巴利森苷E和巴利森苷A)和284 nm(检测橙皮苷)。结果:天麻素、巴利森苷E、巴利森苷A和橙皮苷分别在1.52~30.40μg/mL、1.78~35.60μg/mL、4.16~83.20μg/mL、9.34~186.80μg/mL范围内线性关系良好(r≥0.9993),平均加样回收率分别为98.42%、96.93%、99.49%和97.66%,RSD分别为0.96%、0.67%、1.09%和0.77%。结论:所建立的方法操作便捷、结果准确,可用于小儿葫芦散的质量控制。Objective:To establish an HPLC gradient elution method for simultaneous determination of gastrodin,parishin E,parishin A and hesperidin in Xiaoer Hulu Powder.Methods:The chromatographic separation was carried out on Agilent Extend C18(200 mm×4.6 mm,5μm)column,with mobile phase composed of methanol-0.1%formic acid solution as a mobile phase in gradient elution,and the column temperature was 25℃.The flow rate was 0.8 mL/min,The detection wavelength were 220 nm for gastrodin,parishin E and parishin A,and 284 nm for hesperidin.Results:Gastrodin,parishin E,parishin A and hesperidin showed good linear relationships within the ranges of 1.52-30.40μg/mL,1.78-35.60μg/mL,4.16-83.20μg/mL,9.34-186.80μg/mL(r≥0.9993),whose average recoveries were 98.42%,96.93%,99.49%,97.66%,with the RSD of 0.96%,0.67%,1.09%,0.77%,respectively.Conclusion:The method is simple,accurate,which is favorable to improve overall quality control of Xiaoer Hulu Powder.
