摘  要:目的:探讨补肾活血祛瘀法对早期闭合性骨折模型大鼠愈合程度及血管新生的影响及作用机制。方法:40只大鼠随机分为对照组、模型组、阳性组(麝香接骨胶囊,0.5 g/kg)和试验组(补肾活血祛瘀法,5 g/kg),每组10只。建立闭合性骨折模型,干预后14 d后,观察并比较各组大鼠的血清骨钙素、Ⅰ型胶原的表达、骨折区组织质量以及血液黏度,放射免疫法检测各组大鼠的血浆中血栓素B2(TXB2),6-酮前列腺素F1α(6-Keto-PGF1α)的含量以及TXB2/6-Keto-PGF1α酶联免疫法(ELISA)检测各组大鼠的血清中血管内皮生长因子(VEGF)水平,并对比各组大鼠的骨痂毛细血管数量。取骨组织病理切片观察骨痂微血管生成及骨折愈合情况。结果:与对照组比较,模型组大鼠的骨折面缺损明显形成,即造模成功,Ⅰ型胶原含量、骨钙素含量水平、血清TXB2水平、TXB2/6-Keto-PGF1α水平、全血黏度和血浆黏度明显高于对照组,血清6-Keto-PGF1α水平明显低于对照组(P<0.05)。干预后,阳性组和试验组大鼠缺损毛细血管以及静脉新生,且已有大量成骨细胞的形成,Ⅰ型胶原含量、骨钙素含量以及骨折区组织质量、TXB2和TXB2/6-Keto-PGF1α和血黏度、血浆黏度明显低于模型组,血清6-Keto-PGF1α水平明显低于模型组(P<0.05),而试验组大鼠的骨折面中的骨小梁和骨性骨痂的形成明显,两组间比较均有显著性差异(P<0.05)。结论:补肾活血祛瘀法可促进早期闭合性骨折模型大鼠骨折愈合,降低血液黏度,促使血管新生。Objective:To explore the effect and mechanism of Bushen Huoxue Quyu method on healing degree and angiogenesis of early closed fracture model rats.Methods:40 rats were randomly divided into the control group,the model group,the positive group(Shexiang Jiegu Capsule,0.5 g/kg)and the experimental group(Bushen Huoxue Quyu Formula,5 g/kg),with 10 rats in each group.The expressions of serum osteocalcin,typeⅠcollagen,tissue quality in the fracture area and blood viscosity were observed and compared among the groups after 14 days of intervention.The serum contents of TXB2,6-Keto-PGF1αand TXB2/6-Keto-PGF1αwere detected by radioimmunoassay,ELISA method was applied to detect serum level of VEGF,and the number of epiphyseal capillaries was compared in each group.Also,microangiogenesis and fracture healing of the epiphysis were observed.Results:Compared to the control group,the fracture surface defects were obviously formed in the model group,which showed the successful modeling,the levels of typeⅠcollagen,osteocalcin,TXB2,TXB2/6-Keto-PGF1α,whole blood viscosity and plasma viscosity were significantly higher,and the serum level of 6-Keto-PGF1αwas significantly lower in the model group than those in the control group(P<0.05).After the interventions,angiogenesis of capillaries and veins could be clearly seen,and a large number of osteoblasts had formed in the positive group and the experimental group.TypeⅠcollagen content,osteocalcin content,and tissue quality of the fracture area,TXB2 and TXB2/6-Keto-PGF1α,blood viscosity,and plasma viscosity were significantly lower after interventions compared to those in the model group(P<0.05);serum level of 6-Keto-PGF1αwas significantly lower in the intervention groups than that in the control group(P<0.05).The formations of bone trabecula and bony callus were obvious in the experimental group compared to those in the positive group(P<0.05).Conclusion:Bushen Huoxue Quyu method can promote fracture healing in early closed fracture model rats,reduce blood viscosity and promote a
