摘 要:目的基于网络药理学方法分析半夏泻心汤的关键活性成分治疗糖尿病胃轻瘫(DGP)胃排空延迟的作用机制。方法利用中药系统药理学数据库和分析平台(TCMSP)获取半夏泻心汤的主要活性成分及其对应的作用靶点,在人类基因数据库(GeneCards)中搜索DGP胃排空延迟相关的靶点,利用Cytoscape3.7.1软件构建中药活性成分-靶点网络,运用STRING数据库构建蛋白相互作用(PPI)网络,利用DAVID6.8在线工具进行靶点的GO功能富集和KEGG通路富集分析。结果筛选出半夏泻心汤复方中的152个活性成分,作用于108个靶点,其中关键靶点21个,包括AKT1、ESR1和STAT3等。GO功能分析共获得392条富集结果,其中,生物过程主要涉及对药物的反应,对凋亡、蛋白磷酸化和基因表达的调控等299条;分子功能涉及酶结合、蛋白结合、细胞因子活性等37条;细胞组分涉及细胞质、细胞外区域、线粒体和细胞核等56条。KEGG分析结果显示,TNF、HIF-1和PI3K-Akt等信号通路与半夏泻心汤治疗DGP胃排空延迟密切相关。结论本研究初步揭示半夏泻心汤治疗DGP胃排空延迟是多成分、多靶点和整体调节的复杂过程,为后续研究提供新的理论依据。Objective To explore the core active components and targets of Banxia Xiexin Decoction and the possible mechanism of treating delayed gastric emptying in diabetic gastroparesis based on network pharmacology.Methods Major active components and corresponding target genes of Banxia Xiexin Decoction were obtained by Traditional Chinese Medicine Systems Pharmacology(TCMSP),and the target genes of diabetic gastroparesis were obtained by GeneCards.Cytoscape3.7.1 software was used to construct the active components-target network of TCM.The network of protein interaction(PPI)was constructed by using STRING database.The DAVID6.8 online tool was used for the gene ontology(GO)analysis and kyoto encyctopedia of genes and genomes(KEGG)pathway enrichment analysis of target genes.Results Totally 152 active components of Banxia Xiexin Decoction were selected,which acted on 108 targets,21 of which were key targets,involving AKT1,ESR1 and STAT3.GO functional analysis obtained a total of 392 enrichment results.Among them,the biological process mainly involved 299 items such as response to drugs,regulation of apoptosis,protein phosphorylation and gene expression,etc.;molecular functions involved 37 items such as enzyme binding,protein binding,and cytokine activity;cell components involved 56 items including cytoplasm,extracellular area,mitochondria and nucleus.KEGG results showed that signal pathways such as TNF,HIF-1 and PI3K-Akt were closely related to delayed gastric emptying in diabetic gastroparesis treated by Banxia Xiexin Decoction.Conclusion This study preliminarily reveals that Banxia Xiexin Decoction for delayed gastric emptying in diabetic gastroparesis is a complex process with multiple components,multiple targets and overall regulation,which provides a new theoretical basis for the follow-up studies.
