摘  要:目的探讨熊果酸(ursolic acid,UA)对1型糖尿病(type 1 diabetes,T1DM)大鼠Th1、Th2型转录因子T-bet/GATA-3表达的影响。方法复制T1DM大鼠模型,随机分为空白组、模型组、二甲双胍组和UA组,每天给予100 mg/kg相应药物灌胃,每周监测体质量、空腹血糖,持续6周;免疫组化染色观察胰腺病理变化;ELISA法检测血清胰岛素(insulin,INS)、IL-2、IL-4含量;qRT-PCR法检测脾脏T-bet、GATA-3 mRNA水平;Western blot法检测脾脏T-bet和GATA-3蛋白表达水平。结果与空白组比,模型组空腹血糖、IL-2、T-bet mRNA及蛋白水平、T-bet/GATA-3 mRNA比值均升高(P<0.01),体质量、INS、IL-4、GATA-3 mRNA及蛋白水平均降低(P<0.01);与模型组比,UA组空腹血糖、IL-2、T-bet mRNA及蛋白水平、T-bet/GATA-3 mRNA比值均降低(P<0.01),其体质量、INS、IL-4、GATA-3蛋白水平均升高(P<0.01),GATA-3 mRNA转录水平升高(P<0.05)。结论UA可通过下调T-bet,上调GATA-3的表达纠正Th1/Th2细胞因子失衡,从而发挥降糖作用。Objective To investigate the effects of ursolic acid(UA)on the expression of Th1 and Th2 transcription factors T-bet/GATA-3 in type 1 diabetic(T1DM)rats.Methods T1DM rat models were duplicated and randomly divided into blank group,model group,metformin group,and UA group.The rats were given intra-gastric administration of corresponding drug(100 mg/kg)every day.Body weight and fasting blood glucose were monitored weekly for 6 weeks.Immunohistochemical observation of pancreatic pathology changes;Serum insulin(INS),IL-2,IL-4 levels were detected by ELISA;Spleen T-bet and GATA-3 mRNA levels were detected by qRT-PCR;Spleen T-bet and GATA-3 were detected by Western blot Protein expression level.Results Compared with the blank group,the fasting blood glucose,IL-2,T-bet mRNA and protein levels,and the T-bet/GATA-3 mRNA ratio in the model group increased(P<0.01);Body weight,INS,IL-4,GATA-3 mRNA and protein levels decreased(P<0.01);Compared with the model group,fasting blood glucose,IL-2,T-bet mRNA and protein levels,and T-bet/GATA-3 mRNA ratios in the UA group were all reduced(P<0.01),body weight,INS,IL-4,GATA-3 protein levels increased(P<0.01),the level of GATA-3 mRNA transcription increased(P<0.05).Conclusion UA could correct the Th1/Th2 cytokine imbalance by down-regulating the expression of T-bet and up-regulating GATA-3 expression,thereby exerting hypoglycemic effect.
