摘  要:优化瓶尔小草的多糖的提取工艺及体外抗氧化活性,以瓶尔小草多糖提取量为考核指标考察液料比、超声时间、超声功率和提取温度对其多糖提取量的影响,在单因素基础上结合Box-Behnken响应面分析法优化瓶尔小草多糖的提取工艺,并以清除DPPH自由基和·OH能力评价其多糖的体外抗氧化活性。瓶尔小草多糖的最佳提取工艺为:液料比7mL/g、超声时间43min、超声功率300W和提取温度70℃,此时其多糖的含量为9.82mg/g;瓶尔小草多糖有较强的抗氧化活性,其清除DPPH自由基、·OH的半抑制浓度(IC50)分别为0.029mg/mL和0.454mg/mL,为进一步研究瓶尔小草的药用价值和保健功能提供理论依据。The purpose of the study is to determine the optimal extraction technology of polysaccharides from ophioglossum vulgatum. Effects of material to solvent ratio,ultrasonic time,ultrasonic power and temperature on the extraction of polysaccharides were studied by single factor and response surface methodology. The in vitro antioxidant activity of polysaccharides was evaluated by scavenging DPPH and hydroxyl radicals. The optimal extraction technology was as follow :solvent to material ratio 7 mL/g,ultrasonic time 43 min,ultrasonic power 300 W and temperature 70℃. Under the above conditions,the polysaccharides content of ophioglossum vulgatum was 9.82 mg/g. The polysaccharides of ophioglossum vulgatum showed strong antioxidant activity :the half inhibitory concentration(IC50)for scavenging of DPPH and hydroxyl radicals were 0.029 mg/mL and 0.454 mg/mL,respectively. The study provides theoretical basis for further research on medicine value and dietary supplemental function of ophioglossum vulgatum.
