摘 要:目的实验观察丹参通脉胶囊对Wistar大鼠血小板聚集功能、组织型纤溶酶原激活剂(t-pA)及其抑制物(PAI)活性的影响,探究丹参通脉胶囊对缺血性脑血管疾病的治疗机理。方法于2009年1—4月选取Wistar大鼠48只,随机分为4组,分别为空白对照组、丹参通脉胶囊2 g·kg^(-1)组、丹参通脉胶囊1 g·kg^(-1)组、血塞通片54 mg·kg^(-1)组,每组12只,对比研究各组大鼠的血小板聚集功能;于2009年5—7月选取Wistar大鼠60只,随机分为5组,分别为空白对照组、模型组、丹参通脉胶囊2 g·kg^(-1)组、丹参通脉胶囊1 g·kg^(-1)组、血塞通片54 mg/kg^(-1)组,每组12只,对比研究各组大鼠组织纤溶酶原激活剂(t-PA)及其抑制剂(PAI)的活性。结果运用丹参通脉胶囊的小鼠血小板聚集功能M丹参通脉胶囊2 g·kg^(-1)组为(42.16±16.34),丹参通脉胶囊1 g·kg^(-1)组为(49.58±20.33),均低于空白对照组(69.52±19.77)(P<0.05)。丹参通脉胶囊2 g·kg^(-1)组和丹参通脉胶囊1 g·kg^(-1)的小鼠t-pA活性分别为(2.11±0.62)IU/mL和(1.68±0.53)IU/mL均高于模型组的t-pA活性(0.96±0.37)IU/mL(P<0.001),其PAI活性分别为(0.21±0.11)AU/mL和(0.24±0.13)AU/mL均低于模型组的PAI活性(0.41±0.13)AU/mL(P<0.001)。结论丹参通脉胶囊能明显降低血小板聚集率,抑制血小板聚集功能,并能明显增强t-PA活性、降低PAI活性,且与用药剂量相关。Objective To experimentally observe the effects of Danshen Tongmai Capsules on the platelet aggregation function,tissue-type plasminogen activator(t-pA)and its inhibitor(PAI)activities in Wistar rats,and explore the effects of Danshen Tongmai Capsules on ischemia Therapeutic mechanism of sexual cerebrovascular disease.Methods From January to April 2009,48 Wistar rats were selected and randomly divided into 4 groups,namely the blank control group,the Danshen Tongmai capsule 2 g·kg-1 group,and the Danshen Tongmai capsule 1 g·kg-1 group,Xuesaitong tablet 54 mg·kg-1 group,12 rats in each group,to compare and study the platelet aggregation function of rats in each group;60 Wistar rats were selected from May to July 2009,randomly divided into 5 groups:blank control group,model control group,Danshen Tongmai Capsule 2 g·kg-1 group,Danshen Tongmai Capsule 1 g·kg-1 group,and Xuesaitong Tablet 54 mg·kg-1 group,12 rats in each group,comparative study of the activity of plasminogen activator(t-PA)and its inhibitor(PAI)in each group of rats.Results The platelet aggregation function of mice using Danshen Tongmai Capsules was(42.16±16.34)in the Danshen Tongmai capsule 1 g·kg-1 group and(49.58±20.33)in the Danshen Tongmai capsule 1 g·kg-1 group,which were lower than(69.52±19.77)in the blank control group(P<0.05).The t-pA activity of mice in the Danshen Tongmai capsule 2 g·kg-1 group and Danshen Tongmai capsule 1 g·kg-1 group were(2.11±0.62)IU/mL and(1.68±0.53)IU/mL,respectively,which were higher than those of the model group(0.96±0.37)IU/mL(P<0.001),the PAI activities of(0.21±0.11)AU/mL and(0.24±0.13)AU/mL were lower than those of the model group by(0.41±0.13)AU/mL(P<0.001).Conclusion Danshen Tongmai Capsule can significantly reduce the platelet aggregation rate,inhibit platelet aggregation function,and can significantly enhance the activity of t-PA and reduce the activity of PAI,which is related to the dosage.
