摘 要:目的探究氧化苦参碱对病毒性心肌炎小鼠心肌中B淋巴细胞瘤-2基因(Bcl-2)及Bcl-2相关X蛋白(Bax)表达的影响及意义。方法选取25只健康SD雄性小鼠,其中5只作为正常组,不作处理,其余20只建立病毒性心肌炎模型,并随机分为模型组、药物对照组、低、高剂量组各5只,并对各组分别干预。对各组心功能指标进行检测,酶法速率法检测乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)、肌酸激酶(CK)水平,酶联免疫吸附实验法检测鼠白细胞介素(IL)-8、C反应蛋白(CRP)、肿瘤坏死因子(TNF)-α水平,免疫透射比浊法检测丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平,Western印迹法检测Bcl-2、Bax相对表达量。结果高剂量组左室舒张末期内径(LVEDd)、左室收缩末期内径(LVESd)水平均显著低于药物对照组、低剂量组、模型组,左室射血分数(LVEF)水平高于模型组、药物对照组、低剂量组(P<0.05)。高剂量组LDH、CK-MB、CK、IL-8、CRP、TNF-α、MDA水平、Bax表达均显著低于其他各组(P<0.05)。高剂量组SOD、GSH-Px水平、Bcl-2表达显著高于其他各组(P<0.05)。结论使用高剂量氧化苦参碱对病毒性心肌炎小鼠进行干预,能够改善心功能及心肌酶水平,抑制炎症反应,提升抗氧化能力,调控Bcl-2、Bax表达,减轻心肌受损严重程度。Objective To investigate the effect of oxymatrine on the expression of B lymphoma (Bcl)-2 gene and Bax in myocardium of mice with viral myocarditis and its significance.Methods 25 healthy SD male mice were selected, 5 of which were treated as normal group, and the remaining 20 were established as viral myocarditis models. They were randomly divided into model, drug control, low-dose and high-dose groups, 5 of which were respectively treated. Cardiac function indexes were tested, the rate of enzymatic method was used to detect lactate dehydrogenase (LDH), creatine kinase isoenzyme (CK-MB), creatine kinase (CK) levels, enzyme-linked immunosorbent experiment was used to examine interleukin (IL)-8, C-reactive protein (CRP), tumor necrosis factor (TNF)-α level, the immune transmission turbidimetry was used to detect MDA, SOD, GSH-Px levels, Western blot method was used to detect the Bcl-2 and Bax.Results LVEDd and LVESd in high dose group were lower than those of drug control, low-dose and model groups, LVEF was higher than that of model, drug control and low-dose groups ( P <0.05). The levels of LDH, CK-MB, CK, IL-8, CRP, TNF-α, MDA and Bax in high-dose group were lower than those in other groups ( P <0.05). The levels of SOD, GSH PX and Bcl-2 in high-dose group were higher than those in other groups ( P <0.05).Conclusions Intervention with high dose oxymatrine in mice with viral myocarditis could improve cardiac function and myocardial enzyme level, inhibit inflammation, enhance antioxidant capacity, regulate the expression of Bcl-2 and Bax, and reduce the severity of myocardial damage in mice.
