摘 要:目的探讨硼硅酸盐生物玻璃浸提液对大鼠成骨细胞增殖分化的影响及其相关机制。方法取ROS1728大鼠成骨细胞,采用CCK-8实验及碱性磷酸酶染色分别检测不同浓度浸提液干预下成骨细胞的增殖及活性情况;检测钙盐沉积量;采用RT-PCR法及Western-blot法分别检测不同浓度浸提液干预下成骨细胞Wnt3a、β-catenin、BMP2 mRNA表达及PI3K、p-AKT、RUNX2、beta-actin蛋白表达。结果与对照组比较,高剂量组与低剂量组的成骨细胞增殖能力及活力均明显增高,高剂量组成骨细胞增殖能力明显低于低剂量组(P<0.05),成骨细胞活力明显高于低剂量组(P<0.05);高剂量组与低剂量组成骨细胞钙盐沉积量均明显增高(P<0.05),高剂量组成骨细胞钙盐沉积量均明显高于低剂量组(P<0.05);高剂量组与低剂量组成骨细胞Wnt3a、β-catenin、BMP2 mRNA及PI3K、p-AKT、RUNX2、beta-actin蛋白表达水平均明显增高(P<0.05),高剂量组成骨细胞Wnt3a mRNA及PI3K、RUNX2蛋白表达水平明显高于低剂量组(P<0.05)。结论硼硅酸盐生物玻璃浸提液能够促进大鼠成骨细胞的增殖分化,提高钙盐沉积量,其通过激活Wnt及PI3K/AK信号通路而发挥作用。Objective To discuss the effect of borosilicate biological glass leach liquor on proliferation and differentiation of rat osteoblasts and its mechanism.Methods ROS1728 rat osteoblasts were taken,and the proliferation and activity of osteoblasts were detected by CCK-8 experiment and alkaline phosphatase staining.Calcium salt deposition was detected.The mRNA expression of Wnt3a,β-catenin,BMP2 and protein expression of PI3K,p-AKT,RUNX2,beta-actin in osteoblasts were detected by RT-PCR and Western-blot respectively under the intervention of different concentrations of leach liquor.Results Compared with the control group,the proliferation ability and activity of osteoblasts in the high-dose group and the low-dose group significantly increased,and the proliferation ability of osteoblasts in the high-dose group was significantly lower than that in the low-dose group(P<0.05),the activity of osteoblasts was significantly higher than that in the low-dose group(P<0.05).Calcium salt deposition of osteoblasts significantly increased in the high-dose group and the low-dose group(P<0.05),and the calcium salt deposition of osteoblasts in the high-dose group was significantly higher than that in the low-dose group(P<0.05).The mRNA expression of Wnt3a,β-catenin,BMP2 and protein expression of PI3K,p-AKT,RUNX2,beta-actin in osteoblasts significantly increased in the high-dose group and the low-dose group(P<0.05),and the mRNA expression of Wnt3a and protein expression of PI3K,RUNX2 in the high-dose group was significantly higher than that in the low-dose group(P<0.05).Conclusion Borosilicate biological glass leach liquor can promote the proliferation and differentiation of rat osteoblasts and increase calcium salt deposition by activating Wnt and PI3K/AKT signaling pathways.
