摘  要:目的建立虎杖膏的高效液相色谱法(HPLC)指纹图谱。方法采用HPLC-二极管阵列检测器(DAD)方法,Xtimate C 18(250 mm×4.6 mm,5μm)色谱柱,乙腈-0.1%磷酸水溶液,梯度洗脱;流速1.0 mL/min;检测波长286 nm;柱温30℃;进样量10μL;依据《中药色谱指纹图谱相似度评价系统》(2012版)建立13批虎杖膏的通用模式,然后在模型之间进行相似度的评价。结果创建了虎杖膏HPLC指纹图谱,确定20个共有峰,13批样品的相似度均>0.95,并进行峰归属,指认了虎杖苷、大黄素、芦荟大黄素、白藜芦醇、大黄素甲醚、大黄酸以及大黄素-8-葡萄糖苷7个成分。结论此方法稳定性和重复性良好,对于虎杖膏质量控制有重要的参考价值。Objective To establish the High Performance Liquid Chromatogram(HPLC)fingerprints of Polygonum cuspidatum ointment.Methods HPLC-DAD method was developed to establish fingerprints,Xtimate C 18(250 mm×4.6 mm,5μm)was adopted and eluted in a gradient mode with 0.1%phosphoric acid and acetonitrile as the mobile phase.The column temperature was 30℃;the flow rate of the mobile phase was 1.0 ml/min;the detection wavelength was 286 nm,and the injection volume was 10μl.Similarity evaluation system for chromatographic fingerprint of Traditional Chinese Medicine(2012 version)was used to confirm the common peaks of 13 batches of samples,and then evaluate the similarity.Rusults The common mode for Polygonum cuspidatum ointmen was established;the similarity was greater than 0.95 in 13 batches of Polygonum cuspidatum ointmen;20 common fingerprint peaks were identified.Among them,7 peaks were identified as polydatin,emodin,aloe emodin,resveratrol,rhein and emodin-8-glucoside,respectively.Conclusion This method has desirable stability and repeatability,and can be applied for quality control of Polygonum cuspidatum ointment.
