摘 要:目的:观察艾灸对乳腺增生(MGH)模型大鼠的干预效果,以及对MGH模型大鼠血清雌二醇(E2)、孕激素(P)水平和乳腺组织雌激素受体α(ERα)、孕激素受体(PR)蛋白表达的影响。方法:将40只雌性成熟未孕SD大鼠随机分为空白组、模型组、艾灸组和电针组,每组10只。对模型组、艾灸组和电针组大鼠进行造模,造模成功后,艾灸组和电针组分别进行艾灸和电针干预,空白组和模型组不进行干预。艾灸和电针穴位相同,均分为甲、乙两组,甲组为天宗、肝俞、足三里,乙组为合谷、屋翳、膻中。甲乙两组穴位隔日交替干预,干预5 d,休息2 d为1个疗程,连续干预4个疗程。干预结束后,测量各组大鼠乳头高度和直径,观察各组大鼠乳腺组织形态,并用高通量液相芯片技术检测各组大鼠血清E2、P水平,免疫印迹法检测各组大鼠乳腺组织ERα、PR蛋白表达。结果:干预后,艾灸组和电针组大鼠乳头高度明显低于模型组,乳头直径明显小于模型组(P<0.05);乳腺导管直径均显著缩小,乳腺导管管腔形态相对规则,管腔内脱落组织和分泌物显著减少或消失,乳腺导管上皮细胞数目减少且排列相对规则,乳腺腺泡及乳腺小叶数量均明显减少;血清E2水平明显低于模型组,血清P水平明显高于模型组,乳腺组织ERα、PR蛋白表达明显低于模型组,差异均有统计学意义(P<0.05)。艾灸组和电针组大鼠上述指标比较,差异无统计学意义(P>0.05)。结论:艾灸治疗乳腺增生模型大鼠疗效较好,其机制可能与调节大鼠血清E2、P水平及ERα、PR蛋白表达有关。Objective:To observe the intervention effect of moxibustion on mammary gland hyperplasia(MGH)model rats,and to investigate its effect on the model rats’serum levels of estradiol(E2)and progestogen(P),and on the expression of estrogen receptorα(ERα)and progesterone receptor(PR)protein.Methods:A total of 40 non-pregnant female mature SD rats were randomly divided into blank group,model group,moxibustion group and electroacupuncture group,with 10 rats in each group. Rats in the model group,the moxibustion group and the electroacupuncture group were made into MGH model. After successful modeling,rats in the moxibustion group and the electroacupuncture group were intervened by moxibustion and electroacupuncture respectively,while those in the blank group and the model group were not given any intervention. The moxibustion group and the electroacupuncture group were identical in terms of acupoints. The acupoints were divided into two groups:Tianzong(SI11),Ganshu(BL18) and Zusanli(ST36) in group A,Hegu(LI04),Wuyi(ST15) and Danzhong(RN17) in group B. During intervention,acpoints in group A were alternated with those in group B every other day. One treatment course included 5 intervention days and 2 rest days,totally for 4 courses. After intervention,the height and diameter of the rats’ nipple were measured,the morphology of the rats’ mammary gland tissue was observed,the rats’ serum levels of E2 and P were detected by high-throughput liquid chip technology,and the expressions of ERα and PR protein in the rats’ breast tissue were detected by western blotting method. Results:After intervention,the nipple height and diameter of the rats in the moxibustion group and the electroacupuncture group were significantly lower and smaller than those in the model group(P<0.05). On the morphology of mammary gland tissue,the main changes of the rats in the moxibustion group and the electroacupuncture group were as follows:The diameter of mammary duct was significantly reduced,the shape of mammary duct lumen was relatively
