摘 要:目的通过Box-Behnken响应面法优化萸黄连总生物碱的提取工艺。方法在单因素实验的基础上,以乙醇体积分数、液料比、提取时间和提取温度为影响因素,以总生物碱提取率为评价指标,通过Box-Benhnken响应面法优化提取工艺。结果最佳提取工艺为:乙醇体积分数为70%,液料比为11∶1,提取时间为2.0 h,提取温度为82℃,提取2次,该工艺条件下总生物碱提取率为11.19%。结论Box-Behnken响应面法能较为准确地优选萸黄连总生物碱的提取工艺,该提取工艺方法可靠且精密度高,适合扩大生产。Objective To optimize the exaction technology of the total alkaloids from Coptis chinensis Franch.by Box-Behnken response surface methodology.Methods Based on the results of single factor experiments,taking the volume fraction of ethanol,solid-liquid ratio,extraction time and extraction temperature as the influencing factors,the total alkaloid yield from Coptis chinensis Franch.was the evaluation index.The extraction technology was designed and optimized by the Box-Benhnken methodology.Results The optimal extraction conditions were as follows:the volume fraction of ethanol was 70%,solid-liquid ratio was 11∶1,2.0 h per time,and 82℃heating reflux extraction for 2 times,the total alkaloid yield of this process was 11.19%.Conclusion In this study,the extraction process of the total alkaloids from Coptis chinensis Franch.optimized by Box-Behnken response surface methodology is reliable and highly precise,and is suitable for large-scale production.
