摘  要:目的探讨特应性皮炎(AD)与银屑病之间差异表达的微小RNA(miRNA)及其生物学功能。方法采用第二代测序技术对8例AD及8例银屑病皮损组织的miRNA表达谱进行高通量测序,筛选差异表达的miRNA。使用实时荧光定量PCR技术,选取上、下调表达差异显著的miRNA进行高通量测序结果的验证。针对差异表达miRNA进行靶基因预测,取交集后进行基因本体功能及京都基因和基因组百科全书通路富集分析。结果AD与银屑病相比,有204个差异表达的miRNA,包括168个表达上调的miRNA和36个表达下调的miRNA。上调基因hsa-miRNA-133a-3p和下调基因hsa-miRNA-199b-3p的验证结果与检测结果具有较好的一致性。差异表达miRNA的靶基因多富集在蛋白结合、离子结合、生物调节、细胞代谢等分子功能和生物功能上,多涉及代谢途径、磷脂酰肌醇-3激酶(PI3K)/蛋白激酶B(Akt)信号通路、EB病毒感染及丝裂原活化蛋白激酶信号通路等。结论AD与银屑病之间存在多个差异表达的miRNA,其涉及多种生物功能及通路;PI3K/Akt通路、脂质代谢途径等通路在AD和银屑病的发生及病情演变中发挥重要作用。Objective To explore the microRNAs(miRNAs)with differential expression between atopic dermatitis(AD)and psoriasis and their biological function.Methods High-throughput sequencing of miRNA expression profiles was performed on 8 AD skin lesion samples and 8 psoriasis skin lesion samples using next-generation sequencing technology,then miRNAs with differential expression were screened.The miRNAs with significant differential(up-or down-regulated)expression were selected to verify the results of high-throughput sequencing by using real-time quantitative PCR.The targeted genes of the miRNAs with differential expression were predicted,then Gene Ontology functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes enrichment analysis were performed after interaction for the targeted genes.Results There were 204 miRNAs with differential expression between AD and psoriasis,among which 168 were up-regulated and 36 were down-regulated.The verification results of up-regulated gene hsa-miRNA-133a-3p and down-regulated gene hsa-miRNA-199b-3p were well consistent with the detection results.The targeted genes of the miRNAs with differential expression were enriched in molecular and biological functions including protein binding,ion binding,biological regulation and cell metabolism,and mainly involved in metabolic pathway,phosphatidylinositol 3-kinase(PI3K)/protein kinases B(Akt)signaling pathway,EB virus infection and mitogen-activated protein kinase signaling pathway,ect.Conclusion Multiple miRNAs with differential expression exist between AD and psoriasis,involving various biological functions and pathways;PI3K/Akt pathway,lipid metabolism pathway and other pathways may play an important role in the occurrence and development of AD and psoriasis.
