摘  要:为了研究菠萝洁粉蚧对有机磷杀虫剂在分子水平上的应激反应,采用RNA-seq高通量测序的方法分别对清水处理和杀扑磷药液处理的菠萝洁粉蚧的转录组进行了测序,组装后获得52806个Unigenes,其中15890个Unigene在Nr、SwissProt、KOG和KO四个数据库中有注释。与清水处理的对照组相比,杀扑磷药液处理组中共发现8389个差异表达基因,其中表达量上调的有3068个,下调的有5321个。乙酰胆碱酯酶(AchE)、细胞色素P450酶系(P450s)、羧酸酯酶(CarEs)、谷胱甘肽S-转移酶(GSTs)等相关抗逆、解毒基因表达量以下调为主,这可能与以致死中浓度(LC50)杀扑磷药液浸泡30 s的处理方式有关,即杀扑磷对菠萝洁粉蚧处理剂量相对较高。参与分子功能的基因大多体现在结合和催化活性方面,在脂肪酸代谢、淀粉与蔗糖的代谢、溶酶体、甘油酯代谢、神经活性配体受体相互作用和类固醇生物合成等代谢通路显著富集。To explore the molecular stress reaction of Dysmicoccus brevipes in response to methidathion,transcriptomess of D.brevipes treated with water or methidathion were sequenced and assembled.The sequences were assembled into 52806 unigenes,of which 15890 unigenes were annotated in Nr,SwissProt,KOG and KO transcriptome libraries.Compared with the water treatment,the methidathion treatment was found to have 8389 significant differentially expressed genes(DEGs)in D.brevipes,of which 3068 were up-regulated and 5321 down-regulated.The DEGs related to stress resistance and detoxification such as AchE,P450s,CarEs and GSTs were mainly down-regulated,which might be due to the exposure of D.brevipes to LC50 methidathion for 30 seconds,or treatment with methidathion at a higher rate.The annotated DEGs involved in molecular functions were mainly concentrated on binding and catalytic activity.The DEGs were significantly enriched in metabolism pathways such as fatty acid metabolism,starch and sucrose metabolism,lysosome,glycerolipid metabolism,neuroactive ligandreceptor interaction and steroid biosynthesis.
