摘  要:目的观察染镧仔鼠的皮质神经元细胞变化以及皮质组织内血管内皮生长因子(VEGF)蛋白含量表达的变化,研究VEGF在镧致大鼠神经毒性中的作用。方法 Wistar雌性大鼠32只随机分成对照组及低、中、高剂量氯化镧(LaCl_3)组,每组8只自妊娠开始分别饮用浓度为0.00%、0.25%、0.50%、1.00%的LaCl_3溶液,经血液循环对仔鼠染镧,分娩后以母乳喂养的方式染镧,断乳后仔鼠可自行摄入LaCl_3溶液进行染镧。仔鼠出生后49 d麻醉处死,取脑组织进行尼氏染色,观察仔鼠皮质神经细胞形态变化。以水迷宫空间探索实验,检测仔鼠学习记忆能力的变化。以western-blot法检测VEGF蛋白的表达水平变化。单因素方差法分析组间的差异,SNK法进行组内的两两比较。结果:与对照组相比,LaCl_3组仔鼠皮质中表达尼氏体神经细胞的数量逐渐减少,差异有统计学意义(F=10.465,P<0.001)。在空间探索实验中,LaCl_3组仔鼠进入目标象限的次数和在目标象限内的时间短于对照组,差异有统计学意义(F=3.072,P<0.05)。与对照组相比,LaCl_3组仔鼠皮质神经细胞中VEGF蛋白表达降低,差异有统计学意义(F=133.848,P<0.001)。结论镧暴露使VEGF表达水平下降,降低了对神经细胞的保护作用,导致神经细胞损伤。Objective To observe the changes of cortical neurons and the expression of vascular endothelial growth factor(VEGF)protein in cortical tissues of lanthanum-stained rats,and to study the effect of VEGF on lanthanum-induced neurotoxicity in rats.Methods Thirty-two Wistar female rats were randomly divided into control group and low,medium and high dose lanthanum chloride(LaCl3)groups.From the beginning of pregnancy,eight mice in each group were given LaCl3 at concentrations of 0%,0.25%,0.5%and 1.0%,respectively,and young rats could be dyed with lanthanum through blood circulation.Lanthanum was dyed by breastfeeding after delivery.After weaning,LaClf solution can be ingested by mice to dye lanthanum.The rats were anesthetized and sacrificed 49 d after birth,then the cortical tissues were collected and stained with the Nissl Staining method,and the morphological changes of cortical nerve cells were observed.The experiment of water maze space exploration was used to detect the changes of learning and memory ability of baby mice.The expression level of VEGF protein was detected by Western-blot method.The differences among groups were analyzed by one-way variance method,and the pairwise comparisons were made by SNK method.Results Compared with the control group,the number of Nissl neurons in the cortex of LaCl3 group decreased gradually and the difference was statistically significant(F=10.465,P<0.001).In the space exploration experiment,the mice of the LaCl3 group entered the target quadrant less frequently and spent less time in the target quadrant than the control group,and the difference was statistically significant(F=3.072,P<0.05).Compared with the control group,the expression of VEGF protein in cortical nerve cells of the LaCl3 group was decreased,and the difference was statistically significant(F=133.848,P<0.001).Conclusion Lanthanum chloride exposure decrease the expression of VEGF and the protective effect on nerve cells,and led to nerve cell damage.
