右美托咪定对SNI大鼠的红细胞流变学效应研究

摘  要:目的观察选择性神经损伤(spared nerve injury,SNI)大鼠的红细胞流变学变化以及氧化应激程度,探究右美托咪定对SNI大鼠的治疗作用及其潜在机制。方法纳入30只SD大鼠,随机分为对照组(S组)、模型组(M组)、右美托咪定干预组(D组),每组10只。观察于术前(T0)、术后3天(T1)、术后7天(T2)、术后14天(T3)四个时间点各组大鼠的机械缩足反射阈值(mechanical withdrawl threshold,MWT)及热缩足反射潜伏期(thermal withdrawl threshold,TWL),各时间点的血清氧化应激指标水平脂质过氧化终产物(malonydialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、皮质醇(cortisol,Cor)以及红细胞流变学指标(红细胞聚集指数、红细胞变形指数、红细胞刚性指数)。结果M组大鼠出现痛觉敏化的表现,从T1开始MWT及TWL值相比于S组均显著下降,并表现出过度氧化应激水平(MDA及Cor升高、SOD下降)以及不良红细胞流变学变化(红细胞聚集指数及刚性指数升高、变形指数下降),与S组相比差异有统计学意义(P<0.05);右美托咪定可明显抑制上述变化,D组大鼠MWT及TWL值相比于M组均显著增加,抑制过度的氧化应激状态,使SNI大鼠的MDA及Cor下降、SOD上升;此外,右美托咪定还可改善红细胞血液流变学,使SNI大鼠的红细胞聚集指数及刚性指数下降、变形指数升高(P<0.05)。结论右美托咪定对SNI大鼠具有镇痛作用,并抑制过度的氧化应激且改善红细胞流变学变化。Objective To observe the changes in erythrocyte rheology and the degree of oxidative stress in SNI rats,and explore the therapeutic effect of dexmedetomidine on SNI rats and its potential mechanism.Methods Thirty SD rats were randomly divided into Sham control group(S group),model group(M group)and dexmedetomidine intervention group(D group),n=10.Mechanical withdrawl threshold(MWT)and thermal withdrawl latency(TWL)of all rats were tested before operation(T0),postoperative 3 days(T1),postoperative 7 days(T2),postoperative 14 days(T3).At each time point,the serum levels of oxidative stress indicators(MDA,SOD,Cor)and erythrocyte rheology indicators(erythrocyte aggregation index,erythrocyte deformation index,erythrocyte rigidity index)were detected.Results The rats of M group exhibited a significant pain sensitization.Accordingly,MWT and TWL of rats in M group were significantly decreased compared with S group from T1 to T3(P<0.05).In addition,excessive oxidative stress level and worse erythrocyte rheology change were detected in M group compared with S group(P<0.05);Dexmedetomidine could significantly inhibit the above changes.MWT and TWL of rats in D group were significantly increased compared with those in M group(P<0.05),and the excessive oxidative stress was alleviated by administration of dexmedetomidine.Furthermore,dexmedetomidine also improved erythrocyte rheology change of SNI rats,the erythrocyte aggregation index and rigidity index of D group were decreased compared with M group,meanwhile,deformation index of D group was increased compared with M group(P<0.05).Conclusion Dexmedetomidine has the analgesic effect on SNI rats,and improve its excessive oxidative stress and erythrocyte rheology change.
