摘  要:[目的]建立一种简便快捷且稳定经济的盐酸大观霉素高效液相色谱检测方法。[方法]选用C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-四氢呋喃-水(35∶35∶30),检测波长415 nm,柱温30℃,流速1 mL/min,进样量20μL的高效液相色谱条件,研究该条件下检测样品含量的可行性,并与国家兽药典中的抗生素微生物检定法(管碟法)的检测结果进行对比。[结果]盐酸大观霉素浓度在100~900μg/mL与峰面积呈良好的线性关系,回收率为99.6%,批内变异系数为0.55%,稳定性良好。同一样品2种方法的检测结果相对偏差为0.42%,符合有关规定。[结论]该试验使用的高效液相色谱法(HPLC)较抗生素微生物检定法操作简便且准确度和稳定性更高。[Objective]To establish a simple,rapid,stable and economical high performance liquid chromatography method for the detection of spectinomycin hydrochloride.[Method]C 18 chromatographic column(250 mm×4.6 mm,5μm)was selected;mobile phase:acetonitrile-tetrahydrofuran-water(35∶35∶30),detection wavelength 415 nm,column temperature 30℃,flow rate 1 mL/min,and high performance liquid phase color 20μL.The feasibility of measuring the sample content by this method was studied,and the results were compared with the test results of the antibiotic microorganism assay(cup-plate method)in the National Veterinary Pharmacopoeia.[Result]The concentration of spectinomycin hydrochloride at 100-900μg/mL showed a good linear relationship with the peak area,the recovery rate was 99.6%,the intra batch variation coefficient was 0.55%,and the stability was good.The relative deviation of the results of the two methods of the same sample was 0.42%,which was in conformity with the relevant regulations.[Conclusion]The high performance liquid chromatography(HPLC)used in this experiment is simpler,more accurate and more stable than the antibiotic microbiological assay.
