不同剂量浓度二陈汤对PCOS模型大鼠性激素水平和卵巢形态学的影响

摘　　要：目的观察二陈汤低、中、高不同剂量浓度对多囊卵巢综合征(PCOS)模型大鼠的性激素水平和卵巢形态学变化的影响。方法按体质量将36只SD雌性大鼠随机分为正常组6只和造模组30只,造模组在颈背部皮下注射脱氢表雄酮复制PCOS模型大鼠,正常组在颈背部皮下注射大豆油剂。造模结束后,将造模组大鼠随机分为模型组,二陈汤低、中、高剂量组和二甲双胍组,并以正常组作为对照。二陈汤低、中、高剂量组分别给予二陈汤2.175、4.350、8.700 g/(kg·d)灌胃,二甲双胍组给予二甲双胍0.3 g/(kg·d)灌胃,正常组和模型组给予等体积[10 mL/(kg·d)]灭菌饮用水灌胃。干预4周后,运用酶联免疫吸附法(ELISA)检测各组血清睾酮(T)、卵泡刺激素(FSH)、促黄体生成素(LH)水平,并采用苏木素-伊红(HE)染色法观察各组卵巢组织形态结构。结果与正常组比较,模型组T、LH、FSH水平明显上升(P<0.01);与模型组比较,除二甲双胍组FSH无明显变化外,其余各用药组T、FSH水平明显下降(P<0.01或0.05),二陈汤中剂量组及二甲双胍组LH水平下降(P<0.05),而二陈汤低、高剂量组LH水平有下降趋势,但差异无统计学意义(P>0.05);与正常组比较,模型组卵巢组织镜下可见大量囊状扩张卵泡;与模型组比较,二陈汤低、中剂量组卵巢组织可见各级卵泡,囊状卵泡数量减少,二陈汤高剂量组、二甲双胍组卵巢组织仍可见大量囊状扩张卵泡。结论二陈汤能够显著下调PCOS模型大鼠T、LH、FSH的激素水平及改善卵巢形态学变化,以中剂量二陈汤效果显著。Objective:To observe the effects of different concentrations of Erchen Decoction on the levels of sex hormones and morphological changes of ovaries in polycystic ovary syndrome(PCOS)model rats.Methods:According to body mass,36 female SD rats were randomly divided into the normal group(n=6)and the model group(n=30).PCOS models were induced by subcutaneous injection of dehydroepiandrosterone(DHEA)into the neck,and for the normal group,soybean oil was injected subcutaneously into the neck.After the modeling,the PCOS model rats were randomly divided into model group,Erchen Decoction low,medium and high dose group and metformin group.The low,medium and high dose groups were given Erchen Decoction 2.175,4.35 and 8.7 g/(kg·d)by gavage,the metformin group was given metformin 0.3 g/(kg·d)by gavage,the normal group and the model group were given a equal volume[10 mL/(kg·d)]of sterile drinking water.After 4 weeks of intervention,serum levels of testosterone(T),follicle-stimulating hormone(FSH)and luteinizing hormone(LH)were measured by enzyme-linked immunosorbent assay(ELISA).Hematoxylin-Eosin(HE)staining was used to observe the histological structure of ovary in each group.Results:Compared with the normal group,the levels of T,LH and FSH in the model group were significantly increased(P<0.01).Compared with model group,except that the FSH hormone level of the metformin group had no obvious change,the T,FSH hormone levels of other treatment groups significantly decreased(P<0.01 or 0.05),and the LH hormone levels of the medium dose group and the metformin group decreased significantly(P<0.05),while the LH hormone levels of the low dose and the high dose group had a downward trend(P>0.05).Compared with the normal group,a large number of cystic expanded follicles were observed in the model group.Compared with the model group,all levels of ovarian follicles were observed and the number of cystic follicles reduced in the ovarian tissues of the low and medium dose groups.However,a large number of cystic expanded follicles were
