摘  要:利用线粒体DNA D-loop高变区为标记,研究云南六个小型猪之间的亲缘关系,以期从分子水平揭示它们的遗传多样性及其分布规律,为其遗传资源的保护及开发利用提供参考。采集滇南小耳猪(版纳、盈江、金平、芒市)、迪庆藏猪、明光小耳猪的耳组织,进行DNA提取及扩增、测序、拼接,并利用MEGA软件构建系统树,分析云南六个小型猪之间的亲缘关系。结果表明六个小型猪群中,版纳小耳猪(BN)与迪庆藏猪(DQ)遗传距离最远;版纳小耳猪(BN)、盈江小耳猪(YJ)、金平小耳猪(JP)与芒市小耳猪(MS)聚为一类,其中,盈江小耳猪(YJ)与芒市小耳猪(MS)聚为一小类,版纳小耳猪(BN)与金平小耳猪(JP)聚为一小类;明光小耳猪(MG)与迪庆藏猪(TP)聚为一类。研究证明云南六个小型猪群体遗传距离的大小与其相对地理距离的远近、生活环境及猪种类型吻合。Mitochondrial DNA(mtDNA) D-loop hypervariable region was taken to serve as a marker to study the genetic relationship among six minipigs in Yunnan province,in order to reveal the genetic diversity and distribution of six minipigs in Yunnan province at the molecular level, and provide a reference for their protection of genetic resources.The eav tissues of Diannan small ear pig(Banna、Yingjiang、Jinping、Mangshi),Diqing Tibetan pig,Mingguang small ear pig were collected to extract their DNA and amplification,sequencing and assembly were conducted,in addition,MEGA software was used to construct a phylogenetic tree.The phylogenetic tree showed that Banna small pig(BN) had ulterior relationship with Diqing Tibetan pig(DQ). Banna small ear pig(BN), Yingjiang small ear pig(YJ), Jinping small ear pig(JP) and Mangshi small ear pig(MS) were clustered, among them, YJ and MS pig were in a small cluster, while BN and JP pig in the other one. Mingguang small ear pig(MG) and Diqing Tibetan pig(TP) were clustered. So it is concluded the size of the population genetic distance is consistent with its relative geographical distance, living environment and pig species.
