摘 要:人脐带间充质干细胞是一种多能干细胞,可用于多种疾病的临床治疗。利用从人类脐带分离的间充质干细胞,以及从该间充质干细胞的培养液中分离提取的外泌体,通过与宫颈癌HeLa细胞共培养,研究间充质干细胞及其外泌体对HeLa细胞生物学表型的影响。结果表明:脐带间充质干细胞共培养均极显著抑制HeLa细胞的增殖、迁移及侵袭等表型,并极显著促进HeLa细胞凋亡。脐带间充质干细胞来源的外泌体处理HeLa细胞后,细胞增殖、迁移及侵袭也同样受到极显著抑制,并极显著促进细胞凋亡。这一研究提示脐带间充质干细胞及其外泌体并未在体外培养条件下促进HeLa细胞的生长、迁移,相反对HeLa细胞的表型具有一定的抑制作用。Human umbilical cord mesenchymal stem cell is a type of pluripotent stem cell and it has been reported that it can be used to treat many diseases clinically. In this study, mesenchymal stem cells isolated from human umbilical cord and exosomes extracted from the culture medium of the mesenchymal stem cells were used to be co-cultured with cervical cancer HeLa cells. Cell co-culture technique was used to study the effects of the mesenchymal stem cells and their exosomes on some cell biological phenotypes of HeLa cells. The main results were as follows: Co-culture of umbilical cord mesenchymal stem cells significantly inhibited HeLa cell phenotypes such as cell proliferation(P<0.001), cell migration(P<0.001) and cell invasion(P<0.001), and significantly promoted HeLa cell apoptosis(P<0.01). After the treatment of exosome derived from umbilical cord mesenchymal stem cells, HeLa cell proliferation(P<0.001), migration(P<0.001), and invasion(P<0.001) were also significantly inhibited, while the apoptosis was significantly promoted(P<0.01). Our research suggests that umbilical cord mesenchymal stem cells and their exosomes do not promote HeLa cell growth, migration and other development related phenotypes, on the contrary, it may have a certain inhibitory effect on HeLa cells.
