摘  要:为研究MYL1、MYL2基因在猪的不同组织中的mRNA的表达水平。本试验以大白猪为实验动物,首先,提取其心脏、肝脏、背最长肌、脂肪等组织的总RNA;其次,扩增MYL1、MYL2基因的cDNA序列;最后,以GAPDH基因作为内参,应用实时荧光定量PCR技术检测MYL1、MYL2基因在不同组织中mRNA的表达水平。结果表明:MYL1基因在背最长肌中表达量最高,MYL2基因在心肌和背最长肌表达量最高,在其它器官表达量很少甚至几乎不表达,说明MYL2基因可能对骨骼肌生长和发育起调节作用。本研究表明MYL1和MYL2基因的mRNA在同一猪种不同组织器官间表达量呈现显著性差异,可能与不同组织器官的功能相适应,可为进一步研究猪的肌肉生长发育机理提供理论基础。To study the expression levels of mRNA of the MYL1, MYL2 genes in different tissues of pigs. In this experiment, Large White pigs were used as experimental animals. First, the total RNA of the heart, liver,longissimus dorsi muscle, fat and other tissues was extracted. Secondly, the cDNA sequence of MYL1 and MYL2 genes was amplified. Finally, the GAPDH gene was used as an internal reference. Real-time quantitative PCR was used to detect the mRNA expression levels of MYL1 and MYL2 genes in different tissues. The results showed that the expression of MYL1 gene was highest in the longissimus dorsi muscle, and the expression of MYL2 gene was the highest in the myocardium and the longissimus dorsi muscle. The expression of MYL2 gene was the least in other organs, indicating that the MYL2 gene may be involved in regulating skeletal muscle growth and development. This study showed that the m RNA expression of MYL1 and MYL2 genes showed significant differences in the expression of different tissues and organs of the same pig breed, which may be compatible with the functions of different tissues and organs, which may provide a theoretical basis for the further study of the muscle growth and development mechanism of pigs.
