摘 要:体细胞核移植技术(somatic cell nuclear transfer,SCNT),又称之为体细胞克隆(Somatic cloning),是动物胚胎工程领域一项重要的实验技术手段,对动物种质资源保存、畜牧业发展和生物医学研究等都具有重大意义。目前在大家畜体细胞克隆研究中发现,克隆胚胎发育率普遍低下,如猪由体细胞克隆胚胎移植后产出子代成功率不足1%,极大地限制了体细胞克隆技术在农业和医学上的应用。影响克隆胚胎发育率低下因素很多,如体外培养环境、培养液成分、胚胎自身的质量以及克隆胚胎表观遗传修饰等。本研究主要对表观遗传修饰的概念、类型以及胚胎表观遗传修饰的调控对猪体细胞克隆胚胎发育潜能的影响这些方面展开描述,以期为提升猪体细胞克隆效率相关研究与实践提供理论参考。Somatic cell nuclear transfer(SCNT),also known as Somatic Cloning,is an important experimental technology in animal embryo engineering field,but it is of great significance for the preservation of mammalian germplasm resources and the development of agriculture.However,in the current studies on SCNT of large animals,it has been found that the development rate of cloned embryos is generally low,such as the success rate of offspring generation of pigs cloned from somatic cells after embryo transplantation is less than 1%,which greatly restricts the application of somatic cell cloning technology in agriculture and medicine.There are many studies that affect the low development rate of cloned embryos,such as in vitro culture environment,the culture medium,the quality of embryos themselves and the epigenetic modification of cloned embryos.This paper mainly reviews the concept and types of epigenetic modification and the influence of the regulation of epigenetic modification on the embryonic development potential of pig somatic cell cloning,so as to provide theoretical reference for the research and practice of improving the efficiency of pig somatic cell cloning.
