摘 要:本研究通过自组装短肽探索其在软骨细胞体外三维培养的应用和对兔膝关节软骨损伤的修复作用。通过圆二色谱仪(CD)考察比较加入生长因子前后短肽二级结构的变化;原子力显微镜(AFM)和透射电镜(TEM)检测短肽微观结构;刚果红染色宏观观察短肽自组装情况。将SciobioⅡ形成的纳米纤维网络支架作为软骨细胞体外三维培养的材料,检测在细胞在短肽中的生长及凋亡情况;建立兔膝关节软骨损伤模型,通过HE染色检测软骨损伤修复过程的病理改变。结果显示,在加入生长因子前后,短肽的二级结构未发生显著改变;短肽自组装24 h后可形成纳米支架结构,为细胞的三维培养提供了纳米微环境;人正常软骨细胞可以在短肽形成的三维纳米网络支架结构中生长;动物实验表明短肽可加快兔膝关节软骨损伤修复的进程。我们的研究发现,短肽可用于软骨细胞的三维培养,并对兔膝关节软骨损伤有较好的修复作用。To investigate the effects of self-assembling peptide Sciobio II on the application in three-dimensional culture of cartilage cells in vitro and repairation of rabbit cartilage injury.Circular dichroism(CD)spectrum was applied to compare the secondary structure change of peptide after adding the growth factors;Atomic force microscopy(AFM)and transmission electron microscopy(TEM)were used to examine the microstructure of self-assembling peptide;The self-assembly of short peptide was observed by Congo red staining.The nanofiber scaffold formed by self-assembled peptides was used as the material of three-dimensional culture of chondrocytes in vitro;The model of rabbit cartilage injury was established and the pathological change was detected by HE staining.The result showed that there was no significant difference in the secondary structure of peptide after adding growth factor;Hydrogel scaffold provides a three-dimensional environment for human chondrocytes;Animal experiments indicated that Sciobio II enhanced reparation of rabbit knee cartilage injury.It demonstrate that the self-assembling peptide could applied in the three-dimensional cell culture and tissue repairing in knee cartilage.
