摘  要:为了探讨低氧运动对2型糖尿病(typeⅡdiabetes mellitus,T2DM)大鼠心肌组织NOD样受体家族3(NOD-like receptors,NLRP3)炎症体信号通路的影响。通过高脂饲料结合腹腔注射30 mg/kg的链脲佐菌素(streptozotocin,STZ)建立T2DM模型。建模后,T2DM大鼠进行4周的低氧跑台运动(氧浓度约为14.4%,跑台坡度为0,速度为15 m/min,时间为1 h,每周运动5 d)。此外,对大鼠腹腔注射10 mg/kg的NLRP3炎症体特异性抑制剂MCC950,每周2次,共4周。研究显示,与T2DM组(T2DM)比较,低氧运动组(HE)和低氧运动+MCC950组(HE+MCC950)的总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)和低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)水平均下降,而高密度脂蛋白胆固醇(high density liptein cholesterol,HDL-C)水平升高(p<0.05)。与T2DM组比较,低氧运动组和低氧运动+MCC950组的空腹血糖值(fasting blood glucose,FBG)和胰岛素(insulin,INS)均下降(p<0.05)。低氧运动组的心肌病变较T2DM组减轻,但仍有炎性细胞浸润。低氧运动+MCC950组的心肌形态基本恢复正常。与T2DM组比较,低氧运动组的超氧化物歧化酶(superoxide dismutase,SOD)水平升高,而丙二醛(malondialdehyde,MDA)降低(p<0.05)。与T2DM组比较,低氧运动组的血清白细胞介素-1β(interleukin-1,IL-1β)水平和心肌组织中NLRP3和Caspase-1的蛋白表达水平均降低(p<0.05)。总之,低氧运动可明显改善T2DM大鼠的血脂代谢、提高抗氧化能力、抑制炎症等。低氧运动对2型糖尿病大鼠的心肌保护作用部分归因于其对NLRP3炎症体通路的抑制。In order to explore the effect of hypoxic exercise on the signaling pathway of NLRP3 inflammasome in myocardial tissue of type 2 diabetes(T2 DM)rats.The T2 DM model was established by high fat diet combined with intraperitoneal injection of 30 mg/kg streptozotocin(STZ).After modeling,T2 DM rats performed hypoxic treadmill exercise for 4 weeks(Oxygen concentration is about 14.4%,treadmill slope is 0,speed is 15 m/min,time is 1 h,exercise 5 days per week).In addition,rats were intraperitoneally injected with 10 mg/kg NLRP3 inflammasome specific inhibitor MCC950 twice a week for 4 weeks.Studies showed that compared with the T2 DM group(T2 DM),the total cholesterol(TC),triglyceride(TG)and low-density lipoprotein cholesterol(LDL-C)levels of the hypoxic exercise group(HE)and the hypoxic exercise+MCC950 group(HE+MCC950)decreased,while the high density liptein cholesterol(HDL-C)levels increased(p<0.05).Compared with the T2 DM group,the fasting blood glucose(FBG)and insulin(INS)of the hypoxic exercise group and the hypoxic exercise+MCC950 group decreased(p<0.05).The cardiomyopathy in the hypoxic exercise group was reduced compared with the T2 DM group,but there was still inflammatory cell infiltration.Myocardial morphology in the hypoxic exercise+MCC950 group basically returned to normal.Compared with the T2 DM group,the superoxide dismutase(SOD)level in the hypoxic exercise group increased,while the malondialdehyde(MDA)decreased(p<0.05).Compared with the T2 DM group,the levels of serum interleukin-1β(IL-1β)and protein expression of NLRP3 and Caspase-1 in myocardial tissues in the hypoxic exercise group were reduced(p<0.05).In short,hypoxic exercise can significantly improve the metabolism of blood lipids in T2 DM rats,improve antioxidant capacity,and inhibit inflammation.The protective effect of hypoxia on myocardium in type 2 diabetic rats is partly due to its inhibition of the NLRP3 inflammasome pathway.
