摘  要:为分析辅酶Q10对异丙肾上腺素(ISO)诱导的大鼠心肌缺血损伤的心脏保护作用和机制,本研究将30只SD大鼠随机分为3组,每组10只,分别进行如下处理:正常对照组:对大鼠灌胃14 d橄榄油(2mL/kg)。ISO组:对大鼠灌胃14 d橄榄油(2mL/kg),在第13天和第14天对大鼠腹膜内注射ISO(4 mg/kg)。辅酶Q10组:对大鼠灌胃100 mg·kg^-1·d^-1的辅酶Q10,在第13天和第14天对大鼠腹膜内注射ISO(4 mg/kg)。研究显示,辅酶Q10预处理可显著降低ISO诱导的ST段抬高,降低血清标记酶(AST,LDH,CK和CK-MB)的水平,增强ATPase的活性(p<0.05)。此外,辅酶Q10预处理不仅可以增强心肌中SOD、GPx和CAT的活性,还可以降低MDA水平(p<0.05)。组织病理学分析表明,辅酶Q10可以减轻ISO引起的心肌损伤。辅酶Q10预处理可增加NO和NOS活性并降低ET-1和IMA水平(p<0.05)。辅酶Q10预处理可降低心肌细胞凋亡率和心肌组织中Bax和NF-κBP65的表达,并增加Bcl-2的表达(p<0.05)。本研究证明了辅酶Q10对ISO诱导的大鼠心肌缺血性损伤具有重要的心脏保护作用。辅酶Q10预处理可能通过增强ATPase活性、增强内源性抗氧化剂活性和抑制心肌细胞的凋亡等机制在心肌缺血中发挥心脏保护功能。To analysis the protective effect and mechanism of coenzyme Q10 on isoproterenol(ISO)-induced myocardial ischemia in rats.Thirty SD rats were randomly divided into 3 groups,10 in each group,and treated as follows:normal control group:Rats were fed olive oil(2 m L/kg)for 14 days.ISO group:Rats were fed olive oil(2 mL/kg)for 14 days,and ISO(4 mg/kg)was injected intraperitoneally on the 13 th and 14 th days.Coenzyme Q10 group:Rats were given 100 mg·kg^-1·d^-1 of CoQ10 intragastrically,and ISO(4 mg/kg)was injected intraperitoneally on the 13 th and 14 th days.Studies showed that coenzyme Q10 pretreatment significantly reduced the ST-segment elevation induced by ISO,reduced the levels of serum marker enzymes(AST,LDH,CK,and CK-MB)and enhanced the activity of ATPase(p<0.05).Coenzyme Q10 pretreatment not only enhanced the activity of SOD,GPx and CAT in myocardium,but also reduced the level of MDA(p<0.05).Histopathological analysis showed that coenzyme Q10 reduced ISO-induced myocardial damage.Coenzyme Q10 pretreatment increased NO and NOS activity and decreased ET-1 and IMA levels(p<0.05).Coenzyme Q10 pretreatment reduced the apoptosis rate of myocardial cells and the expression of Bax and NF-κBP65 in myocardial tissue,and increased the expression of Bcl-2(p<0.05).This study demonstrates that Coenzyme Q10 has an important cardioprotective effect on ISOinduced myocardial ischemic injury in rats.Coenzyme Q10 pretreatment may play a cardioprotective role in myocardial ischemia through mechanisms such as enhancing ATPase activity,enhancing endogenous antioxidant activity and inhibiting cardiomyocyte apoptosis.
