摘 要:天然抗性相关巨噬细胞蛋白在植物Cd2+、Fe2+等二价离子吸收和转运及非生物胁迫等过程发挥重要作用。本研究根据苎麻转录组测序中的BnNRAMP1片段序列,克隆了基因的cDNA全长。将BnNRAMP1基因进行生物信息学分析发现,其开放阅读框为1638 bp,编码545个氨基酸,分子量约为59.15 kD,理论等电点为7.54。与川桑(XP_024018653)、桑树(AYW01695)、拟南芥AtNRAMP1(NP_001321606)和AtNRAMP6(OAP169900)同源性氨基酸序列比较发现相似性分别为91.93%、91.19%、79.49%、77.21%。组织表达特异性分析发现苎麻雄花中BnNRAMP1基因的表达量比根、茎、叶、茎尖、雌花等组织中的要高;在镉胁迫处理下根和茎中BnNRAMP1基因在4 h时的相对表达量最高,分别是对照组的2.14和1.41倍,结果表明Bn-%NRAMP1基因能响应镉胁迫。BnNRAMP1基因为今后苎麻在富集和耐受重金属镉方面的育种提供帮助。The natural resistance-related macrophage protein gene plays an important role in the process of divalent ion absorption and transport and abiotic stress of plant iron,cadmium and the like.In the study,the full-length cDNA of the gene was cloned based on the BnNRAMP1 fragment sequence in the ramie transcriptome sequencing.Bioinformatics analysis of BnNRAMP1 gene revealed that the open reading frame was 1638 bp,encoding 545 amino acids with a molecular weight of 59.15 kD and a theoretical isoelectric point of 7.54.The similarities of the deduced amino acid sequence with Morus notabilis(XP024018653),Morus alba(AYW01695),Arabidopsis thaliana AtNRAMP1(NP001321606)and AtNRAMP6(OAP169900),were 91.93%,91.19%,79.49%,and 77.21%,respectively.The tissue expression specificity analysis showed that the expression level of BnNRAMP1 gene in male flowers of ramie was higher than that in roots,stems,leaves,shoot tips and female flowers.Under cadmium stress,the relative expression level of BnNRAMP1 gene was the highest in roots and stems at 4 h,which was 2.14 and 1.41 times higher than that in the control group,respectively.The results indicate that the BnNRAMP1 gene can respond to cadmium stress.The BnNRAMP1 gene is useful for the future breeding of ramie in the enrichment and tolerance of heavy metal cadmium.
