摘　　要：MicroRNAs(miRNAs)在植物生长发育与逆境胁迫应答过程中具有重要作用,通过对大豆gma-mi-%R4359b的分析为其功能研究奠定基础。通过生物信息学对大豆gma-miR4359b的前体序列、启动子的顺式作用元件及靶基因进行分析,并构建了gma-mi R4359b植物表达载体。分析结果表明:大豆gma-miR4359b的前体序列长度为67 bp,成熟序列长度为24 bp。通过Plant-CARE网站分析gma-miR4359b启动子序列发现,在该区域除了存在基本启动子元件CAAT-box、TATA-box和TATA之外,还包括激素调控应答反应作用元件、非生物胁迫应答反应元件、参与光应答元件和伤害反应元件等。通过psRNATarget在线预测了gma-miR4359b的靶基因,推断gma-miR4359b可能参与其靶基因对细胞信号转导、细胞调控、翻译过程和非生物胁迫的调控。构建了包含gma-mi R4359b前体序列的gma-miR4359b-pCAMBIA3301植物表达载体。大豆gma-mi R4359b可能参与非生物胁迫响应,在植物生长发育过程中与逆境胁迫应答过程中起到重要作用。MicroRNAs(miRNAs)play an important role in plant growth and development and stress response.The analysis of soybean gma-miR4359 b lays a foundation for its functional studies.The precursor sequence of soybean gma-miR4359 b,the cis-acting element of the promoter and the target gene were analyzed by bioinformatics method,and the gma-miR4359 b plant expression vector was constructed.The results showed that the precursor sequence of soybean gma-mi R4359 b was 67 bp in length and the mature sequence was 24 bp in length.The gma-miR4359 b promoter sequence was analyzed by the Plant-CARE website,and in addition to the basic promoter elements CAAT-box,TATA-box,and TATA,the hormone regulatory response element and the abiotic stress response element were included in the region.Participate in optical response components and damage response components.The target gene of gma-miR4359 b was predicted online by psRNATarget,and it was concluded that gma-miR4359 b may be involved in the regulation of cell signal transduction,cell regulation,translation process and abiotic stress by its target gene.gma-miR4359 b-pCAMBIA3301 plant expression vector containing the gma-mi-%R4359 b precursor sequence was constructed.Soybean gma-mi R4359 b may be involved in abiotic stress response and play an important role in plant growth and development and stress response.
