摘  要:为探究^60Co-γ射线辐射对盐胁迫下杂交桑树幼苗的影响,以杂交桑品种桂桑优12号为试验材料,对种子进行不同剂量^60Co-γ辐射后,调查100 mmol/L NaCl胁迫下萌发幼苗的生长情况,测定叶片中的光合色素含量,叶片与根中的超氧化物歧化酶(SOD)和过氧化物酶(POD)活性、丙二醛(MDA)含量、可溶性蛋白含量和脯氨酸含量以及液泡型Na+/H+逆向转运蛋白基因MnNHX1、过氧化物酶基因POD1、超氧化物歧化酶基因sodC的表达水平。结果表明:200 Gy^60Co-γ辐照杂交桑种子能显著增加盐胁迫下杂交桑幼苗的株高、主根长、地上部分和根部的鲜质量,提高叶片中叶绿素a、叶绿素b、总叶绿素和类胡萝卜素含量,提高叶片和根中SOD和POD活性,降低MDA含量,增加可溶性蛋白和脯氨酸含量,上调MnNHX1、POD1和sodC的表达量,增强杂交桑幼苗的耐盐能力。研究结果为杂交桑在盐碱地的开发利用以及桑树栽培和育种提供了参考。To investigate the effects of^60Co-γradiation on hybrid mulberry seedling under salt stress,the hybrid mulberry variety Guisangyou 12 was used as experimental material.After the seeds were irradiated with different doses of^60Co-γ,plant growth,content of photosynthesis pigment in leaf,superoxide dismutase(SOD)and peroxidase(POD)activity,malonaldehyde(MDA)content,contents of soluble protein and proline in the leaf and root of hybrid mulberry seedling under 100 mmol/L NaCl stress were measured.Moreover,expression levels of Na+/H+antiporter gene MnNHX1 in vacuolar membrane,peroxidase gene POD1 and superoxide dismutase gene sodC were detected.The results showed that 200 Gy^60Co-γradiation could significantly increase the plant height,main root length,fresh weight of shoot and root,contents of chlorophyll a,chlorophyll b,total chlorophyll and carotenoid in leaf,increase SOD and POD activity in leaf and root,decrease MDA content,increase contents of soluble protein and proline,and up-regulate the expression of MnNHX1、POD1 and sodC.Then,salt damage to hybrid mulberry seedling could be alleviated.Results of this study provided references for the development and utilization of saline-alkali soil and for the mulberry cultivation and breeding.
