摘  要:为了探讨胶原代谢与奶牛不孕症的关系,选取中国荷斯坦奶牛20头,其中健康奶牛10头为对照组,屡配不孕奶牛10头为试验组,采用免疫组化方法分析子宫内膜胶原的表达特征,荧光定量PCR方法分析基质金属蛋白酶-2(MMP-2)、MMP-9、PGR、ESR mRNA的相对表达量。结果表明:试验组子宫内膜Ⅰ型胶原、Ⅳ型胶原的表达量均不显著升高(P<0.01);试验组MMP-2、MMP-9、PGR mRNA的表达量均显著低于对照组(P<0.05)。表明屡配不孕奶牛子宫内膜MMP-2、MMP-9 mRNA的低表达,使MMP-2、MMP-9对胶原的降解减少,导致胚泡发育不良及着床失败,子宫内膜孕酮受体的低表达可能是MMP-2、MMP-9低表达的影响因素之一。In order to explore the relationship between collagen metabolism and infertility in dairy cows,20 Holstein cows were selected,including 10 healthy cows as control group and 10 repeated infertile cows as experimental group.Immunohistochemical method was used to analyze the expression characteristics of collagen in endometrium,and real time PCR was used to analyze the relative expression of MMP-2,MMP-9,PGR and ESR mRNA.The results showed that the expressions of type I collagen and type IV collagen in endometrium were higher than those in control group(P<0.01),and the expressions of MMP-2,MMP-9 and PGR mRNA in experimental group were significantly lower than those in control group(P<0.05).These results indicated that the low expression of MMP-2 and MMP-9 mRNA in the endometrium of multiple infertile cows reduced the degradation of collagen by MMP-2 and MMP-9,resulting in dysplasia of blastocysts and implantation failure.The low expression of progesterone receptor in the endometrium may be one of the factors affecting the low expression of MMP-2 and MMP-9.
