高效液相色谱法测定喉蛾散中盐酸小檗碱含量

摘　　要：目的建立测定喉蛾散中盐酸小檗碱含量的高效液相色谱法。方法色谱柱采用Waters Xridge RShield RP18柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.05%磷酸溶液(B),梯度洗脱,柱温为30℃,检测波长为265 nm,流速为1 mL/min,进样量为5μL。结果盐酸小檗碱质量浓度在7.18~143.60μg/m L范围内与峰面积线性关系良好,R2=0.999 9(n=5),平均回收率为99.52%,RSD为1.65%(n=6)。结论该方法操作简便,结果准确,重复性良好,可用于喉蛾散的质量控制。Objective To establish a high performance liquid chromatography( HPLC) method to determine the content of berberine hydrochloride in Hou’e Powder.Methods Waters Xridge Rshield RP18 column( 250 mm × 4.6 mm,5 μm) was used,the mobile phase was acetonitrile(A)-0.05% phosphoric acid solution( B),gradient elution,the column temperature was 30 ℃,the detection wavelength was 265 nm,the flow rate was 1 mL/min,and the injection volume was 5 μL.Results Berberine hydrochloride showed good linear relation with the peak area in the range of 7.18-143.60 μg/mL,R2= 0.999 9( n = 5).The average recovery was 99.52%,RSD was 1.65%( n = 6).Conclusion The method is simple,accurate,accurate and reproducible,which can used for the quality control of Hou’e Powder.
