摘　　要：目的探讨紫云英苷对人结肠癌SW480细胞的诱导凋亡作用及其相关的分子机制。方法利用CCK-8实验检测紫云英苷对SW480细胞的增殖抑制作用,利用流式细胞术检测紫云英苷对SW480细胞的诱导凋亡作用和线粒体膜电位变化情况,利用Western blotting检测细胞凋亡相关蛋白和信号通路相关蛋白的表达。结果CCK-8实验结果表明紫云英苷能够有效抑制SW480细胞的增殖,其效果优于5-FU,且对正常细胞的毒副作用低于5-FU;流式细胞术结果显示紫云英苷能够通过降低线粒体膜电位水平,有效诱导SW480细胞凋亡;Western blotting实验结果表示,紫云英苷能够上调p-p38的水平并降低p-STAT3的水平。结论紫云英苷能够有效抑制SW480细胞发生的增殖情况,通过抑制线粒体的膜电位水平,有效激活线粒体具有的依赖性质,促使该细胞逐渐发生凋亡情况,其调节机制是通过激活p38信号通路并抑制STAT3通路来实现的。Objective To investigate the apoptosis induced by astragalin in human colon cancer SW480 cells and its related molecular mechanism.Methods The inhibitory effect of astragalin on the proliferation of SW480 cells was detected by CCK-8 experiment.The apoptosis induced by astragalin and the changes of mitochondrial membrane potential were detected by flow cytometry.The expressions of apoptosis-related protein and signal pathway-related protein were detected by Western blotting.Results CCK-8 experimental results showed that astragalin could effectively inhibit the proliferation of SW480 cells,and its effect was excellent.The results of flow cytometry showed that astragalin could induce the apoptosis of SW480 cells effectively by decreasing the mitochondrial membrane potential level.The results of Western blotting showed that astragalin could up regulate p-p38 level and down regulate p-STAT3 level.Conclusion Astragalin can effectively inhibit the proliferation of SW480 cells,effectively activate the dependent nature of mitochondria by inhibiting the level of mitochondrial membrane potential,promote the gradual apoptosis of the cell,its regulatory mechanism is achieved through the activation of p38 signal pathway and the inhibition of STAT3 pathway.
