摘 要:目的探讨西格列汀对糖尿病(DM)大鼠血清GLP-1分泌及糖代谢的影响。方法将30只大鼠分为健康组、DM组及抑制剂组,每组10只。DM组及抑制剂组大鼠采用链脲佐菌素建立糖尿病大鼠模型,抑制剂组大鼠利用西格列汀进行灌胃,其他两组采用同体积的生理盐水灌胃,观察三组大鼠血糖、血脂、胰腺形态、胰腺细胞凋亡、GLP-1及DPP-4表达水平。结果DM组大鼠空腹血糖、空腹胰岛素及糖化血红蛋白水平均高于健康组(P<0.05),抑制剂组与DM组相比上述指标水平下降(P<0.05)。与健康组相比,DM组大鼠HDL-C水平下降,TC、TG及LDL-C水平升高(P<0.05);与DM组相比,抑制剂组TC、TG及LDL-C水平降低,HDL-C水平升高(P<0.05)。与健康组相比,DM组血清中GLP水平降低,DPP4水平升高(P<0.05);与DM组相比,抑制剂组血清中GLP-1水平升高,DPP4水平降低(P<0.05)。健康组大鼠胰腺结构完整,腺泡完好,形态层次分明,DM组大鼠胰腺结构错杂,细胞骤减,可见明显纤维增生,炎症浸润;与DM组相比,抑制剂组胰腺形态改善。与健康组相比,DM组胰腺细胞凋亡程度增加(P<0.05),与DM组相比,抑制剂组细胞凋亡程度下降(P<0.05)。与健康组相比,DM组大鼠肠肌间神经丛中GLP-1蛋白显著降低(P<0.05);与DM组相比,抑制剂组GLP-1蛋白表达升高(P<0.05)。结论西格列汀能够改善糖尿病大鼠糖代谢,促进GLP-1表达,抑制DPP-4表达。Objective To investigate the effect of sitagliptin on serum GLP-1 secretion and glucose metabolism in diabetic(DM)rats.Methods Thirty rats were divided into healthy group,DM group and inhibitor group,with 10 rats in each group.Rats in DM group and inhibitor group were given streptozotocin to establish a diabetic rat model.Rats in inhibitor group were given sitagliptin by gavage,and the rats in the other two groups were given the same volume of normal saline.Blood glucose,blood lipids,pancreas morphology,pancreatic cell apoptosis,GLP-1 and DPP-4 expression levels were measured and compared.Results Fasting blood glucose,fasting insulin,and glycosylated hemoglobin levels in DM group were higher than those in healthy group(P<0.05).Compared with healthy group,the HDL-C level was decreased in DM group,while the TC,TG and LDL-C levels were increased(P<0.05).Compared with DM group,the levels of TC,TG and LDL-C were decreased in inhibitor group,while the level of HDL-C was increased(all P<0.05).Compared with healthy group,the serum GLP level was decreased in DM group,while the DPP4 level was increased(P<0.05).Compared with DM group,the serum GLP-1 level in inhibitor group was increased,while the DPP4 level was decreased(all P<0.05).The structure of the pancreas of the rats was complete in healthy group,the acini were intact,and the morphology was distinct.The structure of the pancreas was disordered in DM group,the cells were suddenly reduced,and there was obvious fibrosis and inflammation infiltration.Compared with DM group,the pancreatic morphology was improved in inhibitor group.Compared with healthy group,the degree of pancreatic cell apoptosis was increased in DM group(P<0.05).Compared with DM group,the degree of apoptosis decreased in inhibitor group(P<0.05).Compared with healthy group,the GLP-1 protein in the intestinal myenteric plexus was significantly reduced in DM group(P<0.05).Compared with DM group,the expression of GLP-1 protein in inhibitor group was increased(P<0.05).Conclusion Sitagliptin can improve the
