摘  要:目的:研究健脑益智胶囊(JNYZ)对老年小鼠学习记忆能力的药理作用及其作用机制。方法:将40只老年小鼠,随机分为4组:老年组、JNYZ低、中、高剂量组(2.5,5.0,10.0 g·kg^-1),另设10只4月龄小鼠作为青年对照组。连续灌胃给予健脑益智胶囊治疗60 d。治疗结束后采用Morris水迷宫法测试小鼠的学习记忆能力,免疫组化法检测海马组织中Bax的表达,透射电镜观察神经细胞及其突触的超微结构,生化法和放射免疫法分别测定脑组织中的乙酰胆碱(Ach)含量、胆碱酯酶(AChE)活性和胆碱乙酰转移酶(ChAT)活性,免疫印迹法分析大脑组织中突触素的表达水平。结果:与老年组相比,JNYZ 3个剂量组均可明显提高老年小鼠学习记忆能力(P<0.05,P<0.01,P<0.01),显著升高治疗组大脑组织中的乙酰胆碱含量、ChAT活性(P<0.01,P<0.01,P<0.01)和降低脑组织AChE活性(P<0.05或P<0.01),并能明显减少海马神经细胞中Bax的蛋白表达水平,对老年小鼠大脑神经细胞和突触结构有明显的保护作用,并能有效地提高突触素在大脑组织中的表达水平。结论:JNYZ能够改善和提高老年小鼠的学习记忆能力,可能与它抑制神经细胞凋亡、增强中枢神经系统神经递质和改善神经细胞超微结构等作用有密切的相关性。OBJECTIVE To study the effects of JNYZ on learning and memory in aging mice and discuss its possible mechanisms.METHODS Forty aged mice were randomly divided into 4 groups:aged group,JNYZ low,medium and high dose group(2.5,5.0,10.0 g·kg^-1).Ten mice aged 4 months were involved as young mouse control group.Afterwards,the mice of treatment groups were intragastric administered for 60 days.The learning and memory of mice were tested by Morris water maze.The expression of Bax in hippocampus was measured by immunohistochemistry.Transmission ultra structures of neurons and synapses were observed by electron microscope.The content of acetylcholine(Ach),activities of acetylcholinesterase(AchE)and choline acetyltransferase(ChAT)in brain tissues were measured by biochemical technique and radioimmunoassay.The expression level of synaptophysin in the brain tissue was detected by Western blotting.RESULTS Compared with the aged group,JNYZ could obviously improve the learning and memory ability of aged mice(P<0.05,P<0.01,P<0.01),significantly increase the acetylcholine content and ChAT activity in the brain tissue of the treatment group(P<0.01,P<0.01,P<0.01)and decrease the AChE activity in the brain tissue(P<0.05 or P<0.01),significantly reduce the protein expression level of Bax in the hippocampal nerve cells,significantly protect the brain nerve cells and synaptic structure of aged mice,and effectively increase the expression level of synaptophysin in the brain tissue.CONCLUSION JNYZ can improve the learning and memory ability of aged mice,which may be closely related to its inhibition of neuronal apoptosis,enhancement of neurotransmitters in the central nervous system and improvement of neuronal ultrastructure.
