摘  要:目的建立四逆汤活性成分在血液和心脏部位的微透析采样方法。方法以四逆汤中苯甲酰新乌头原碱、6-姜酚和甘草苷的回收率为指标,采用浓度差法(增、减量法)考察灌流液流速对回收率的影响,并综合实际因素确定四逆汤体内微透析采样流速;采用增量法考察探针外液温度和浓度对回收率的影响;用减量法验证各指标成分在体内10 h的释放率稳定性。结果在不同流速下,各成分在血液和线性探针中的增量法回收率与减量法释放率基本一致,且随着流速的增大而减小,结合采样体积和实验时间,确定以1.0μL·min^-1为体内实验流速;温度对血液探针回收率无显著影响,但在线性探针中,回收率与温度呈现相同趋势;浓度对2种探针各成分的回收率有一定的影响;在2种探针中,各指标成分的体内回收率均能在10 h内维持稳定。结论所建立的微透析方法能够用于研究四逆汤在大鼠血液和心脏部位活性成分的动态变化情况。Objective To establish the method of microdialysis in blood and heart for active components of Sini Decoction.Methods Concentration difference method(incremental and reduction method) was applied and the recovery rate of benzoylmesaconine,6-zingerol and glycyrrhizin in Sini Decoction was used as indexes to investigate the impact of perfusion fluid flow rate on the recovery.The flow rate was determined according to the recovery rate and the actual factors.The incremental method was applied in the in vitro study to investigate impact of probe external fluid temperature and concentration to the recovery rate.Reduction method was applied in the in vivo study to verify the recovery rate stabilities of the index components for 10 hours.Results At different flow rates,the incremental recovery rates of each component in the blood and linear probe were basically the same as the reduction method,and decreased with the increase of flow rate.Considering the sampling volume and the experimental time,a flow rate of 1.0 μL·min^-1 was chosen.The probe external fluid temperature has no effect on the recovery rate of blood probes;but in linear probes,the recovery rate and temperature show the opposite trend.Concentration had small effect on the recovery of each component of the two probes.The reduction method maintained good stability within 10 hours in vivo.Conclusion The established microdialysis method can be used to study the dynamic changes of Sini Decoction in rat blood and heart.
