摘 要:采用酶转化法和金属离子催化法联用制备了人参皂苷Rh2系列异构体,研究了原二醇类人参皂苷(PPD)的酶转化反应条件和非水相中Fe3+水解人参皂苷F2的过程,进行反应条件的优化。结果表明,在乙醇体积分数10%、底物质量分数2.5%,反应温度40℃、反应时间、1.5 h时,酶转化PPD的产物得率最高。对金属离子催化人参皂苷F2的条件进行优化,在乙醇体积分数50%、Fe3+溶液反应浓度0.8 mol/L、底物质量分数1.3%、反应温度50℃、反应时间17 h时,人参皂苷Rh2的得率最高。在最佳条件下,20 g PPD最终转化成9.41 g Rh2,得率为50.6%。反应产物粗品中包括20(S,R)-Rh2、Rh3、Rk2。The ginsenoside Rh2 series isomers were prepared by enzymatic transformation and metal ions catalysis.The enzymatic conversion reaction conditions of ginsenoside PPD and the hydrolysis process of ginsenoside F2 by Fe3+in non-aqueous phase were studied,and the reaction conditions were optimized.The results showed that the yield of PPD could reach highest under the conditions of ethanol concentration 10%,substrate content 2.5%,40℃for 1.5 h.The conditions of ginsenoside F2 catalyzed by metal ions were optimized.The result showed that the highest yield of ginsenoside Rh2 was obtained at the ethanol concentration of 50%,Fe3+concentration of 0.8 mol/L,substrate content of 1.3%,50℃for 17 h.Under the optimal conditions,20 g PPD was converted into 9.41 g Rh2 and the yield was 50.6%,including 20(S,R)-Rh2,Rh3 and Rk2.
