摘 要:目的通过网络药理学研究气血和胶囊治疗气滞血瘀证的活性成分和作用机制。方法借助TCMSP、TCM-MESH等数据库结合文献挖掘的方法,以OB≥30%、DL≥0.18和Caco-2≥-0.4作为标准筛选气血和胶囊的活性成分。基于TCMSP、TCM-MESH、CTD等数据库预测活性成分的靶标,并通过Uniprot和GeneCards数据库根据靶标分子功能将靶标归属到气滞血瘀证的相应病理过程中。采用Cytoscape构建"药物-成分-靶标-病理过程"网络,然后对各网络模块靶标进行GO富集和KEGG通路分析(设定阈值P<0.01)。结果气血和胶囊中185个活性成分、51个靶标蛋白涉及调控血液流变学异常;184个活性成分、68个靶标蛋白涉及改善凝血功能异常;174个活性成分、24个靶标蛋白涉及改善疼痛;191个活性成分、29个靶标蛋白涉及参与炎症;105个活性成分、27个靶标蛋白涉及调节免疫。结论气血和胶囊可通过同时改善血液流变学异常、凝血功能异常、疼痛、炎症及免疫来治疗气滞血瘀证。本研究揭示了气血和胶囊治疗气滞血瘀证的潜在活性成分及机制,为其药效物质基础和作用机制的实验研究提供了依据。Objective To explore the active components and mechanisms of Qixuehe capsule(QXH)in treating qi stagnation and blood stasis syndrome.Methods By the combination of TCMSP,TCM-MESH databases and literature mining,taking OB≥30%,DL≥0.18 and Caco-2≥-0.4 as criteria,the active components of QXH were screened.The target proteins of QXH were predicted based on TCMSP,TCM-MESH,CTD and other databases,and all target proteins were assigned to the corresponding pathological processes of qi stagnation and blood stasis syndrome according to the molecular function of target proteins by Uniprot and GeneCards databases.The"drug-component-target-pathological process"network module of QXH was constructed by Cytoscape.Then,GO enrichment and pathway analysis(P<0.01)were carried out on the targets of each network module.Results Through network pharmacology,QXH may improve hemorheological abnormality mainly via 185 components-51 targets;184 components-68 targets associated with QXH in improving coagulation disorders;174 components-24 targets associated with QXH in relieving pain;191 components-24 targets associated with QXH in antiinflammation;and 105 components-27 targets associated with QXH in improving immunity.Conclusion QXH may regulate and improve the 5 pathological processes such as hemorheological abnormality,coagulation disorders,pain,inflammation and immunity to treat qi stagnation and blood stasis syndrome.This study systematically revealed the potential active components and possible mechanisms of QXH in treating qi stagnation and blood stasis syndrome,which provided a theoretical basis for further experimental research on the chemical material basis and mechanism of action.
