摘 要:为确定天津某猪场病猪死亡原因,以及了解分离到的菌株致病性及耐药性,试验通过采集病猪脑组织、肺组织及关节液进行细菌分离培养及生化特性鉴定,并通过PCR方法鉴定其血清型及毒力基因,对其中血清型毒力基因cps9H进行测序分析,并构建系统发育树,对分离的毒株进行小鼠致病性试验及药敏试验。结果显示,从病死猪体内分离到cps9H型猪源链球菌,与GenBank中登录的序列号为CP002007(中国重庆)的菌株同源性高达99%,毒力基因型为SrtA+mrp+orf2+和gapdh+,小鼠致病性试验表明菌株为强毒株。药敏试验结果表明,菌株对氨苄西林、阿莫西林、头孢曲松、头孢哌酮和头孢噻肟高度敏感;对新霉素、四环素、甲硝唑和庆大霉素等耐药。研究结果表明,本次疫病由猪链球菌9型引起。试验结果为天津地区9型链球菌的诊断和防控提供了试验基础。In order to determine the cause of the death in a pig farm in Tianjin,and to understand the pathogenicity and drug resistance of the isolated strains,in this experiment,bacterial isolation,culture and biochemical characterization were carried out by collecting brain tissue,lung tissue and joint fluid of diseased pigs,and the serotypes and virulence genes were identified by PCR.The serotype virulence gene cps 9H was sequenced and analyzed,and phylogenetic tree was constructed,the pathogenicity and drug sensitivity tests of the isolated strains were conducted in mice.The results showed that Streptococcus suis cps9H was isolated from dead pigs.The homology with the strain CP002007(Chongqing,China)registered in GenBank was 99%.The virulence genotype was SrtA+mrp+orf2+gapdh+.The pathogenicity test in mice determined that the strain was a virulent strain.The drug sensitivity test showed that the strain was highly sensitive to ampicillin,amoxicillin,ceftriaxone,cefoperazone and cefotaxime,and resistant to neomycin,tetracycline,metronidazole,tetracycline and gentamicin.The results showed that the disease was caused by Streptococcus suis type 9.This experiment provided experimental basis for the diagnosis and control of Streptococcus type 9 in Tianjin.
