摘  要:目的研究糖尿病大鼠动物模型中血管内皮细胞生长因子(VEGF)在早期糖尿病视网膜病变(DR)中的变化。方法应用链脲佐菌素(STZ)制备糖尿病大鼠动物模型,采用HE染色、免疫荧光、免疫荧光双重标记技术,分别以胶质纤维酸性蛋白(GFAP)抗体、VEGF抗体为一抗;羊抗兔IgCy3、马抗鼠IgG-FITC为二抗进行免疫荧光染色、免疫荧光双重标记技术。结果在糖尿病大鼠动物模型中,在早期DR中,视网膜厚度随时间延长逐渐增加;VEGF在对照组中主要表达在神经纤维层,实验组中视网膜各层均有表达;主要表达在Müller细胞当中。结论糖尿病大鼠模型中,视网膜随病程延长,损害逐渐加重;各层均产生VEGF,以Müller细胞表达为主。Objective To study the changes of vascular endothelial growth factor(VEGF) in early diabetic retinopathy(DR) in diabetic rats. Methods Diabetic rat model was established by streptozotocin(STZ). HE staining,immunofluorescence and immunofluorescence double labeling techniques were used to detect glial fibrillar acidic protein(GFAP) antibody and VEGF antibody respectively. Sheep anti-rabbit Ig-Cy3 and horse antimouse IgG-FITC were used for immunofluorescence staining and immunofluorescence double labeling technique.Results In the animal model of diabetic rats,the retinal thickness increased gradually with the prolongation of time in the early DR. VEGF was mainly expressed in the nerve fiber layer in the control group,and expressed in all layers of the retinal in the experimental group,especially in Müller cells. Conclusion In diabetic rat model,the retinal damage is aggravated with the prolongation of the course of disease,and the expression of VEGF is dominated by Müller cells in all layers.
