摘 要:目的:观察加味二陈汤对肥胖SD幼鼠脂质代谢和法尼酯衍生物X受体(FXR)的影响。方法:8只妊娠中期大鼠,待产。产出幼鼠后,剔除雌鼠,按每只母鼠8只幼鼠的喂养密度分笼哺乳。正常组母鼠按普通饲料喂养哺乳,仔鼠为正常组。高脂组母鼠按高脂饲料喂养,仔鼠为模型组。21日龄后,模型组幼鼠按高脂饲料继续喂养1周,构建肥胖SD幼鼠模型。将模型组幼鼠随机分为高脂组、辛伐他汀组和二陈汤组。正常组、高脂组均予生理盐水灌胃,辛伐他组和二陈汤组分别以辛伐他汀、加味二陈汤灌胃给药,6周后处死。观察幼鼠体质量和肝脏质量,全自动生化分析仪观察脂质代谢情况,PCR法测定TNF-α和FXR表达量。结果:与正常组比较,高脂组幼鼠的体质量和肝脏质量均明显升高(P<0.01),脂质代谢中的TG、TC、LDL-C均升高(P<0.05),HDL-C降低(P<0.05),肝脏TNF-α和FXR均升高(P<0.05)。与高脂组比较,二陈汤组大鼠的血清LDL-C和肝脏FXR明显降低(P<0.01),肝脏TNF-α降低(P<0.05)。结论:加味二陈汤能够降幼龄SD肥胖大鼠体质量和肝脏质量,改善大鼠脂质代谢,能够抑制肥胖幼鼠肝脏TNF-α和FXR表达量。Objective To observe the effects of modified Erchen decoction on lipid and farnesoid X receptor(FXR) in obese young SD rats. Methods: Eight mid-pregnancy rats were expected to be delivered. After the young rats were born, the female young rats were removed, and the remaining young rats were divided into cages to suckle at the feeding density of 8 young rats per mother rat. In the normal group, the mother rats were fed with common feed and their baby rats were in the normal group. The rats in high fat group were fed with high fat diet. After 21 days of age, the young rats in the model group were fed high-fat diet for one week to construct an obese SD young mouse model. The young rats in the model group were randomly divided into high-fat group, simvastatin group and Erchen decoction group. Normal group and high-fat group were given normal saline by gavage, simvastatin group and Erchen decoction group were gavaged with simvastatin and modified Erchen decoction respectively for 6 weeks and then killed. The body weight and liver weight of the young rats were observed. The lipid metabolism was observed by automatic biochemical analyzer. The expression of TNF-αand FXR was determined by PCR. Results: Compared with the normal group, the body weight and liver weight of the young rats in the high-fat group were significantly increased(P <0.01), TG, TC, and LDL-C in lipid metabolism were increased(P <0.05), HDL-C was decreased(P <0.05), and liver TNF-α and FXR were increased(P<0.05). Compared with the high-fat group, the serum LDL-C and liver FXR of rats in the Erchen decoction group were significantly decreased(P<0.01), and liver TNF-α was decreased(P<0.05). Conclusion: The modified Erchen decoction can reduce the body weight and liver weight of young SD obese rats, improve the lipid metabolism of rats, and inhibit the expression of TNF-α and FXR in the liver of obese young rats.
