摘  要:目的:运用网络药理学方法和技术研究"半夏-苍术-香附"组药治疗多囊卵巢综合征(PCOS)的主要活性成分、靶点和通路。方法:通过数据挖掘获得PCOS痰湿型核心组药半夏-苍术-香附;检索中药系统药理学分析平台(TCMSP),筛选出半夏、苍术和香附的主要活性成分和潜在靶点;查询NCBI数据库获得PCOS疾病靶点,找到药物和疾病的共靶点;通过David数据库和Omicshare数据库对共靶点进行基因功能GO富集分析和KEGG富集分析;采用Cytoscape软件构建中药-成分-靶标网络。结果:从"半夏-苍术-香附"组药中筛选得到40个候选活性分子,作用于681个靶点,其中与PCOS重合的23个疾病靶点主要涉及癌症通路、流体剪切应力与动脉粥样硬化通路、HIF-1信号通路、糖尿病并发症中的AGE-RAGE信号通路等生物学过程中的多条信号通路。结论:"半夏-苍术-香附"组药通过槲皮素、山奈酚、β-谷甾醇、木犀草素、汉黄芩素等活性物质形成多靶点、多通路调控网络,增强免疫机制,改善胰岛素抵抗,降低炎症细胞因子水平,达到系统治疗PCOS的目的。Objective: Using network pharmacological methods and technologies to study the main active ingredients, targets and pathways of the Banxia-Cangzhu-Xiangfu group drugs for the treatment of polycystic ovary syndrome(PCOS). Methods: The PCOS phlegm-dampness core group drug Banxia-Cangzhu-Xiangfu was obtained through data mining. The main active ingredients and potential targets of Banxia, Cangzhu and Xiangfu were searched by searching the Chinese Medicine System Pharmacological Analysis Platform(TCMSP). The database obtains PCOS disease targets, finds common targets of drugs and diseases, performs gene function GO enrichment analysis and KEGG enrichment analysis of the common targets through David database and Omicshare database. Cytoscape software is used to construct a Chinese medicine-component-target network. Results: 40 candidate active molecules were screened from the Banxia-Cangzhu-Xiangfu group drugs, which acted on 681 targets, of which 23 disease targets coincided with PCOS mainly involved cancer pathways, fluid shear stress and Multiple signaling pathways in biological processes such as atherosclerosis pathway, HIF-1 signaling pathway, and AGE-RAGE signaling pathway in diabetes complications. Conclusion: Banxia-Cangzhu-Xiangfu group medicines form a multi-target, multi-path regulatory network through quercetin, kaempferol, β-sitosterol, luteolin,and baicalein to enhance the immune mechanism and insulin resist, reduce the level of inflammatory cytokines,and achieve the goal of systemic treatment of PCOS.
